PDCD4 and PTEN were examined by Western blot and their genes also detected by qRT-PCR in order to find the function of miR-21.Results: Proliferation of U87 cell was inhibited and apoptosis was induced by anti-miR-21.The mRNA level of PDCD4 and PTEN did not change significantly after cell transfection compared to the control. The protein level of PDCD4 and PTEN increased after inhibition of miR-21.Conclusions: It suggested miR-21 an oncogene role which was vestified by experiment that proliferation of U87 cell inhibited and apoptosis induced by Anti-miR-21. Because the protein level of PDCD4 and PTEN increased while ever their mRNA level not changed significantly when transfected with the anti-mir-21 ,it mention us miR-21 maybe play a post-transcriptional character to several target genes of glioma . It maybe a new way and a promising molecular target for us to treat glioma in future. | | Keywords/Search Tags: | microRNA, miR-21, glioma, apoptosis, target gene, gene therapy | PDF Full Text Request | Related items |
