The Effect Of Normal Saline With Different Temperature On The Spinal Cord During The Spinal Operation

Objective: To investigate the effect of normal saline (NS) with different temperature on morphological changes and nitric oxide synthase (NOS) expression of spinal cord,with the aim of choosing suitable temperature NS to flush on the spinal cord during the spinal operation.Methods: 160 SD adult rats receiving operation opening spinal cord take the T9 as center. Among these rats,40 in the test group received flushing of the 37℃NS , 40 in the test group received flushing of the 20℃NS , 40 in the test group received flushing of the 4℃NS ,while the other 40 in the control group only received spinal cord operation without any flushing. Resumed the spinal canal after one hour. Take the spinal cord in the one hour,24 hour and 7days. Observed the change from the light microscope and the electronic microscope.Moreover, take the spinal cord in the 24 hour. Observed the NOS positive neuron amount was measured byβ-NADPH histochemistry. Observed the content of water in the rat spinal cord after operation was measured by dry-wet method. In the control group and the 37℃test group ,the spinal cord have not evidence change while in the 20℃and 4℃test group,the spinal cord have evidence injury. The averious kinds changes in the three time points ,the changes are most obviously in the 24 hour. However, the changes of the spinal cord recoverd after seven days. b. The content of water in the spinal cord was( 66.53±0.61)% in control group, (66.75±1.00)% in 37℃group,(70.55±0.77)% in 20℃group,(71.92±2.50)% in 4℃group. The spinal cord of control group and 37℃group contained less water than those of 20℃group and 4℃group. c. The amount of NOS positive neuron of the spinal cord was 18.75±2.12, 18.63±1.41, 14.75±1.67, 8.13±1.25 in control, 37℃, 20℃and 4℃group, respectively. The control group and 37℃group showed more NOS positive neuron than those of 20℃group and 4℃group.Conclusions:The temperature of the NS flushing the spinal cord in the operation will affect the spinal cord,4℃,20℃NS have the injury to the spinal cord,but 37℃NS have not. It is suitable to choose 37℃NS for flushing on the brain and spinal card during the operation.
