ObjectiveTo investigate the effect of curcumin on inhibiting the proliferation of lung cancer ,then study the neucleoproteinβ-catenin,the key factor of Wnt signaling pathway, to explore the possible molecular mechanism of inhibiting the proliferation of lung cancer cells.Methods1. A549 cells were traditionally cultured in vitro and intervened by different concentrations of curcumin. After 48 hours,MTT assay was employed to evaluate the effect on the proliferative inhibition of A549 cells,and the relationship between drug concentration and proliferation inhibition rate was analyzed.2. The percentage of cells in G0/G1 phase was examined by the flow cytometry, for analyzing the effect of curcumin on cell cycle.3. After the intervention of curcumin, A549 cell nuclearβ-catenin protein content was assayed by Westem blot,and c-myc gene expression was examined by RT-PCR,to explore the molecular pharmacological mechanisms of curcumin.Result1. MTT assay showed A549 cells proliferation inhibition rate had an upward trend after the 48 hours intervene of different concentrations of curcumin. The more concentrations,the greater inhibitory rate. And a dose-response relationship between them can been seen(P <0.05).2. The flow cytometry analysis showed that after the intervention of curcumin in A549 cells, the percentage in G0/G1 phase were significantly more than the control group without curcumin to deal with(P <0.05).3. The result of Westen blot and RT-PCR indicated that the intracellular core protein expression ofβ-catenin levels in the experimental group cells were significantly lower than the negative control group(P <0.05).ConclusionsCurcumin can prevent A549 lung cancer cells to enter S phase from G1 phase,and inhibite the proliferation of A549 cells. Its molecular mechanism may be that through the reduction of nuclearβ-catenin, curcumin can block the Wnt signal transduction pathway and inhibit the expression of c-myc gene,a downstream target gene, to prevent lung cancer cells to enter S phase from G1 phase.
