Objective: Objective to explore the effect of jiaweifeifu decoction on tumor bearing mice with adenocarcinoma of lung A549 cells and Wnt / β-catenin signaling pathway.Methods:(1)The BALB/c-nu mice were divided into 6 groups with 10 mice in each group.Blank control group: free feeding;control group: distilled water by gavage;jiaweifeifu decoction low dose group group: 0.87 g/ml by gavage;jiaweifeifu decoction medium dose group: 1.74 g/ml by gavage;jiaweifeifu decoction high dose group: 3.48 g/ml by gavage;positive group: Cisplatin 4 mg/Kg(0.4 mg/m L)by intraperitoneal injection.After 14 days,the tumor volume and weight of tumor bearing mice were compared to observe the effect of different concentrations of jiaweifeifu decoction on the growth of tumor bearing mice with lung adenocarcinoma A549 cells;(2)Western blot method was used to detect the effect of different tumor groups on the content of total protein and nuclear protein of β –Catenin;(3)The qPCR method was used to detect the expression of C-myc,c-jun,cyclin D1 gene in different groups.Results:(1)Compared with the control group,the tumor growth rate of jiaweifeifu decoction medium dose group,jiaweifeifu decoction high dose group and positive group was slower;jiaweifeifu decoction medium dose group,jiaweifeifu decoction high dose group and positive group could effectively inhibit the tumor growth of A549 tumor bearing mice(P < 0.05),while jiaweifeifu decoction low dose group had no significant inhibitory effect on the tumor growth of A549 tumor bearing mice(P > 0.05),and the difference was not statistically significant.(2)Western blot method showed that compared with the control group,the jiaweifeifu decoction medium dose group,high dose group and cisplatin positive group could inhibit the expression of total protein of β-Catenin(P < 0.05);the jiaweifeifu decoction low dose group had no significant inhibitory effect on the expression of total protein of β-Catenin(P > 0.05),and the difference was not statistically significant;The jiaweifeifu decoction low,medium,high dose group and cisplatin positive groups could reduce the concentration of β – Catenin nucleoprotein(P < 0.05).(3)QPCR method showed that,compared with the control group,the expression of c-myc,c-jun and cyclin D1 could be down regulated in the jiaweifeifu decoction high dose group and the positive group(P < 0.05);the expression of c-myc,c-jun and cyclin D1 was not down regulated in the jiaweifeifu decoction low and medium dose groups,and the difference was not statistically significant(P > 0.05).Conclusion: jiaweifeifu decoction can inhibit the growth of adenocarcinoma of lung A549 tumor bearing mice,and the higher the concentration is,the more obvious the inhibitory effect is;this effect may be achieved through Wnt / β-catenin signaling pathway.
