| Objective:This experiment is in order to examine the differences its between the health of its population and gastric cancer patients,different clinical TNM stages of gastric cancer patients,and different clinicalpathological characteristics of gastric cancer by detecting the health of populations and preoperative patients with gastric cancer peripheral blood in T cells, dendritic cell precursor cells (pDC), and the assessment of immune status in patients with gastric cancer clinical significance. By detecting tumor infiltrating dendritic cells to explore the differences its in different clinicopathological features,different clinical TNM stages of gastric cancer patients of gastric cancer patients and the assessment of local immune function in patients with gastric cancer the clinical significance of state.Methods:( 1 ),Surgical resection of gastric cancer specimens are biopsied by conventional HE staining, to clear organization type, pathological features (grade, depth of invasion, lymph node metastasis, etc.) and clinical stage of it.(2),Infiltrating dendritic cells in gastric cancer tissue tumor are detectde by immunohistochemical method, semi-quantitative analysis of dendritic cell infiltration.(3),T-cell subsets (CD3+,CD4+,CD8+ T cells) of healthy population and preoperative peripheral blood of patients with gastric cancer are analyse by flow cytometric,dendritic cell subsets of precursor cells (pDC,pDC1,pDC2) situation of it are analyse by four-color fluorescence flow cytometry .(4),The measured experimental data are analyse by SPSS13.0 package on a computer,count data usingχ2 test,testing the standard of a = 0.05;Measurement data to test results ( x±s)% indicated that the SPSS13.0 software analysis of the data (cell content) in line with normal distribution, comparison between groups using t test, testing the standard of a = 0.05.Results:(1),Immunohistochemistry results showed that serosal tumor invasion group tumor tissue infiltrating dendritic cells lower than serosal invasion non-group; tumor tissue infiltrating dendritic cells in lymph node metastasis group lower than the group without lymph node metastasis; TNMâ…¢,â…£stage group lower than TNMâ… ,â…¡stage group, the difference was statistically significant (All P values<0.05); formation of different degree of differentiation between gastric cancer group,the difference was no significant difference (P> 0.05).(2),Flow cytometry results showed that peripheral blood of patients with gastric cancer study group CD3+,CD4+ T cells,CD4+/CD8+ ratio was significantly lower than the healthy control group, the difference has statistical significance (All P values <0.05),and between the two groups of CD8+ T cells, differences no significant(P>0.05); Serosal invasion tumor group CD3+,CD4+ T cells,CD4+/CD8+ ratio was significantly lower than no serosal invasion group, the difference has statistical significance (All P value<0.05);tumor lymph node metastasis group CD3+,CD4+ T cells,CD4+/CD8+ ratio was significantly lower than tumor-free lymph node metastasis group, the difference has statistical significance (All P value<0.05);TNMâ…¢,â…£period group CD3+,CD4+ T cells,CD4+/CD8+ ratio was significantly lower than TNMâ… ,â…¡stage group, the difference was statistically significant(All P values<0.05); Different degree of differentiation of gastric cancer group, CD3+,CD4+,CD8+T cells,CD4+/CD8+ ratio of the difference was not statistically significant(All P values>0.05),Different groups of peripheral blood CD8+ T cells was no significant difference (All P value> 0.05).(3),Flow cytometry showed that 40 cases of gastric cancer patients,5 cases of lack of peripheral blood pDC,and the remaining 35 cases of peripheral blood of patients with pDC1/pDC2 ratio (0.751±0.137)lower than in healthy population control group(1.788±0.108)(P < 0.05);pDC(0.369±0.106)lower than in healthy population control group (0.701±0.089)(P < 0.05),there is statistically significant difference; different degree of differentiation of gastric cancer group, no tumor invasion serosal layer and serosal layer invasion group,no lymph node metastasis and metastasis,TNMâ… ,â…¡stage group and TNMâ…¢,â…£period groups in peripheral blood pDC,pDC1/pDC2 ratio of the difference was not statistically significant (All P values>0.05). Conclusions:To assess the status of local immune function in patients with gastric cancer have clinical significance by detecting tumor tissue infiltrating dendritic cells in patients with gastric cancer;To assess the immune status of gastric cancer patients by detecting preoperative peripheral blood T cell subsets (CD3+,CD4+,CD8+ T cells, CD4+/CD8+ ratio), dendritic cell subsets (pDC, pDC1, pDC2, pDC1/pDC2 ratio)of gastric cancer patients have clinical significance,also to enhance tumor immune therapy provides a theoretical basis. |
