Objective:To detect the expression of Maspin in the nomal normal endometrium tissue, atypical hyperplasia of endometrium,and endometrioid adenocarcinoma tissues,to investigate the possible relationship between Maspin expression and clinicalpa- thological parameters in endometrioid adenocarcinoma,and evaluate its possible significant role in tumor igenesis and progression of endometrioid adenocarcinoma.Methods:A immunohistochemical method of Elivision TM plus was used to detect Maspin in 67 cases endometrioid adenocarcinoma,30 cases of atypical hyperplasia of endometrium,and 18 cases of normal endometrium tissue. To analyze the relationship to age,clinicalstage,histological grade,muscular invasion,vascular invasion and lymphnode metastasis.Results:1.The positive expression rates of Maspin protein in normal endometrium, atypical hyperplasia of endometrium, and endometrioid adenocarcinoma tissue were 16.67% (3/18),40.00% (12/30),68.66% (46/67),there was significant difference that the positive expression rates of Maspin protein in the three samples(x~2=18.17,P =0.000,P<0.05);2.From normal endometrium to atypical hyperplasia of endometrium, and endometrioid adenocarcinoma tissue, the positive expression rates of Maspin protein show escalating trend(P<0.05);3.The positive expression rates of Maspin protein most were cytoplasmic staining was26.67%(4/15) in normal endometrium and atypical hyperplasia of endometrium tissue,but 73.91%(34/46)in endometrioid adenocarcinoma,from the data, there was significant difference(x~2=10.75,P =0.001,P<0.05).4.In 67 cases of endometrioid adenocarcinoma,the positive expression rate were 80.00%(20/25)in the age sample of less than or equal to 50 years, 61.91% (26/42)in the age sample of older than 50 years, there was no significant difference (P=0.130,P>0.05);83.33%(25/30)in stageâ… ,68.42%(13/19)in stageâ…¡,44.44%(8/18)in stageâ…¢,compared to the three samples, there was significant difference(x~2=9.063,P=0.011,P<0.05);75%(15/20)in G1,70.97%(22/31)in G2,56.25%(9/16)in G3, compared to the three samples, there was no significant difference(x~2=0.716,P=0.699,P>0.05);73.17%(30/41)in muscular invasion sample of less than 1/2, 61.53%(16/26)in muscular invasion sample of greater than 1/2, compared to the two samples, there was no significant difference(P=0.195,P>0.05);42.86%(6/14) in vascular invasion, 75.47%(40/53)in no vascular invasion, compared to the two samples, there was significant difference(P=0.042,P<0.05);25.00%(2/8)in positive lymph node metastasis, 74.58%(44/59)in negative lymph node metastasis, compared to the two samples, there was significant difference(P=0.006,P<0.05).Conclusion:1. There is low-regulated expression of Maspin in normal endometrium and atypical hyperplasia of endometrium,and up-regulated expression of Maspin in endometrioid adenocarcinoma, but following the progression of carcinoma, the expression was down-regulated, it was suggested that Maspin may play a complicating regulation role in the tumor igenesis and progression of endometrioid adenocarcinoma.2. The positive expression of maspin protein most were nucleus staining in normal endometrium tissue and atypical hyperplasia of endometrium, but, in endometrioid adenocarcinoma, they were most cytoplasmic staining.Nucleolus immuno-expression of Maspin may sign optimum, but, cyto plasmic immun o- expression may play a role in the malignant transformation state of end ometrial tissue.3. Maspin expression showed positive association with the clinical stage,vascular invasion and lymph node metastasis,by contrast, there were no correlated with ages, histological grade and muscular invasion.4.The positive expression of Maspin might have inhibitory effect on invasion and metastasis of endometrioid adenocarcinoma and acts as an effactive and objective marker to indicate the patholological behavior of endometrioid adenocarcinoma.
