| Objective:To explore the relationship between intrahepatic expression of inducible nitric oxide synthase (iNOS)mRNA and nuclear factor-κB (NF-κB ) activation in liver injury in rats with severe acute pancreatitis(SAP)and explore the effect of NAC for reducing liver injury.Methods: Fifty-four SD rats were randomized into sham operation group, SAP group and therapeutic group with N-acetylcysteine (NAC).SAP was inflicted with 4% sodium taurocholate by the method of retrograde pancreatic injection. In the therapeutic group rats recieived one intravenous injection of NAC (200mg/kg) 1h after taurocholate injection. All the animals were sacrificed randomly in 3h,6h,12h after induction of panceatitis. Activation of NF-κB in liver tissue was determined with immunohistochemistry. Intrahepatic expression of iNOS mRNA was assayed by RT-PCR.Meanwhile plasma level of amylase, liver function (ALT, AST) and malondialdehyde (MDA) in serum.Result:SAP model group after the NF-kB was significantly increased, iNOSmRNA over-expression in liver tissue and serum MDA increased. SAP group at different time points significantly higher than the SO group (P <0.05), NAC group was significantly lower than the SAP (P <0.05), NAC Group amylase, AST, ALT and MDA levels compared with the SAP group significantly decreased (P <0.05).Conclusions:The expression of iNOSmRNA and activation of NF-κB are associated with liver injury in SAP. N-acetyl-L-cysteine to some extent, anti- oxidants inhibit the NF-κB activation and thus inhibit the abnormal overexpression of iNOS mRNA. |
