| Objective: To make the rat model of severe SAP, to record the pathological changes of i NOS, pancreas and kidney tissue, and to explore the role of i NOS in the pathogenesis of SAP.Methods: Male Wistar rats(SPF) of a total of 300, were randomly divided according to altitude: Xi’an, Xining, and the Xinghai, the Wenquan, each elevation group were randomly divided into five groups according to treatment, namely: sham group, SAP1 h, SAP6 h, SAP12 h, SAP24 h. Sodium taurocholate in rat pancreatic capsule at multiple points injection approach artificial rat model of severe acute pancreatitis, the sham group, laparotomy after turning pancreas abdomen was closed. At 12 h after successful model rats were sacrificed the sham treatment; respectively in 1h, 6h, 12 h, 24 h after modeling SAP rats were sacrificed success. Find the abdominal aorta to extract each rats arterial 3ml, serum amylase, creatinine and urea nitrogen levels measured by automatic biochemical analyzer immunity; application of surgical techniques to take the kidney tissue, pancreatic tissue, in 30 minutes for B formaldehyde solution to fixed, paraffinembedded pathological score in tissue sections, after HE staining. Applications Image Pro Plus software calculates absorbance measurement of the concentration of i NOS protein in pancreatic tissue immunohistochemistry. Statistical Methods: SPSS 19.0 for analysis of experimental data. Mean ± standard deviation statistics describe the experimental data. ANOVA comparison group data; or Spearman correlation analysis using Pearson correlation analysis of experimental data. Test level α = 0.05.Results: Severe acute pancreatitis rat model successfully established, the sham group amylase, urea nitrogen and creatinine were no significantly increased pancreatic biopsy is also no significant pathological changes; SAP modules made visible amylase, blood urea nitrogen, creatinine increased their pancreatic tissue pathological changes with the time prolonging; 1 hour, 6 hours, 12 hours, 24 hours group which followed elevation, pancreas, kidney damage is more serious. Sham operation group and the artificial production of severe acute pancreatitis, serum amylase, i NOS absorbance, pancreatic injury, renal damage are low, they all have statistically significant difference(P <0.05); each altitude group, pairwise comparison between SAP AMY, renal pathological lesion score, i NOS expression levels follow the growth time is increased expression or damage increased, and the difference was statistically significant(P <0.05); serum amylase at the same time, i NOS expression levels, kidney and pancreas injury with increasing altitude and the expression becomes more or aggravate their differences were statistically significant(P <0.05); each group of i NOS expression and renal pathology scores were positively correlated. Conclusions: 1) at the same time, with the increase of altitude, SAP kidney injury is aggravated trends; 2) i NOS is in severe acute pancreatitis inflammation of kidney dysfunction qualitative indicators, is also quantifiable indicators. |
