| [OBJECTIVE]The aim of study is to optimize the extraction process of Baihuasandi and to prepare a better effective and convenient drug.[METHODS]In this Study,Baihuasandi was extracted by 7 methods.Seven extracts were prepared for comparison of chemical constituents and pharmacological activity.This research was divided into two parts as follow.1.The extraction methods were studied,with the nitidine chloride content and quercetin content determined by HPLC and dry extract yielding rate as index.2.The model,known as dimethylbenzene-induced mouse auris-swell model was duplicated to compare the effect of inflammation. 216 healthy mice were selected and divided into 18 groups at random, 12 per each.For the control group and the test group,the average value of the swelling level of them at different time were collected and analyzed.[RESULTS]Dry extract yielding rate were reduced after ethanol precipitation. Most extracts showed similar chemical profiles for quercetin.Ethanolic extracts were characterized by much more higher nitidine chloride as compared to water extracts.In the experiments of the dimethylbenzene-induced mouse auris-swell model experiment,some anti-inflammation efficacy existed in the all test groups.Furthermore,the high dosage group for Mingled decoction and the high dosage group for Mingled decoction and precipation by 70%alcohol had obvious anti-inflammation efficacy. And the anti-inflammation efficacy was in non-significant difference(p>0.05)[CONCLUSION]In this study,the results of chemical analysis is inconsistent with the pharmacological activity.Anti-inflammatory activity of water extract was investigated through its significant inhibitory effect on the dimethylbenzene-induced mouse auris-swell model.Extraction method is the key to chemical profile and pharmacological activity of Baihuasandi samples.Researchers should take this into consideration when either designing studies or comparing results among studies,as this may explain some of the results seen in the literature.Furthermore,a new idea was provided for the extraction of TCM that we must pay attention to the integration of chemical indexes with pharmacodynamic index. |
