| OBJECTIVE: To assess and compare the expression of heat shock protein glycoprotein 96 (HSPgp96) in gastric cancer and paracancer tissue through cellular level and molecular level research, and thus evaluate its meaning in the diagnosis of gastric cancer and determine its relation to biological behaviors of gastric carcinoma. Besides, the corelation of HSPgp96 to the development of gastric cancer and the underlying mechnism, its value in the prognosis of gastric cancer will also be discussed.METHODS: Immunohistochemistery method and semi-quantitative reverse transcription- polymerasechain reaction(RT-PCR) were applied respectively to detect the expression of HSPgp96 mRNA and protein in 37 cases of gastric cancer (including cancer and paracancer tissue). The clinical and pathological data were combined to analyse the relationship between HSPgp96 expression and the development, differentiation degree, pathological pattern, clinical staging, lymphnode metastasis of gastric cancer.RESULTS: Immunohistochemistery test revealed that Positive expression of HSPgp96 in gastric cancer and paracancer tissue were 75.7% and 29.7% respectively(p<0.01). The expressions in cancer were positive to strongly positive(++~+++), while it was negative to normally positive in paracancer tissue(- ~ +). Positive HSPgp96 expression in gastric cancer were obviously higher than that in paracancer tissue ; HSPgp96 positive expression in poorly differentiated cancer type were higher than that in moderately and well differentiated type(94.4%vs57.9%,P<0.05). The expression rate in positive lymph nodes were higher than that of negative lymph nodes(90.9%vs53.3%, P<0.05). The level of HSPgp96 expression were also correlated with the elevation of TNM classification, and the rates in advantaged stages (III, IV)were significantly higher than those in early stages( I, II) (88.9%vs40.0%, P<0.05). No relationships were showed between HSPgp96 expression and the morphology and depth of invasion of gastric cancer(p>0.05).RT-PCR showed that HSPgp96 mRNA was overexpressed in both gastric cancer and paracancer tissues, and expression level of the former were significantly higher than those of the latter (1.64±0.47 vs 0.81±0.34, P < 0. 01). The expression level of HSPgp96 mRNA in poorly differentiated cancer were (1.71±0.19), higher than that in moderately and well differentiated cancer (1.31±0.14). The expression level in gastric cancer with lymphnode metastesis were higher than that without lymphnode metastesis(1.37±0.09 vs0.95±0.13, P<0.01). Besides, the level was also elevated with the TNM staging advanced stage tumor(III, IV) than ones in early stage tumor( I, II) (1.47±0.23 vs 0.90±0.13, P<0.05). No meaningful results were observed between HSPgp96mRNA calculation and the morphology and depth of invasion of gastric cancer(p>0.05).CONCLUSIONS: HSPgp96 expression in gastric cancer were higher than those in paracancer tissue. The expression of HSPgp96 were closely related to the biological behaviors of gastric carcinoma, such as cancer classification, tumor differentiation and lymph node metastase. HSPgp96 expression can be used as one of the prospective factors. |
