Objective To understand the effects on the mixture of paraphenylenediamine and H2O2 in inducing BALB/c mice to develop lupus-like symptoms.Methods Twenty-one female BALB/c mice were randomly divided into three groups of high PPD(n=7, 800mg/kg), low PPD(n=7, 400mg/kg) and control group(n=7, saline), cutaneous exposure. The level of serum ANA,anti-dsDNA antibody and urinary protein content were detected. 12 weeks after exposure, kidneys were excised to observe kidney pathology examination by HE,PAS staining and electron microscope.Results Since the fourth week, the level of ANA in high PPD and low PPD groups was significantly higher than the control group, and revealed statistical significance (P<0.01).Compared the ANA level of the three groups in the different time point, the differences were statistically significant (P<0.001).Since the fourth and sixth week, the level of ANA in high PPD and low PPD groups was significantly higher than the control group, and revealed statistical significance (P<0.01).Compared the anti-dsDNA antibody levels of the three groups in the different time point, the differences were statistically significant (P<0.01).And compared the urinary Protein content of the three groups since the sixth week, the differences were statistically significant(P<0.05).Light microscope indicated glomerulonephritis(Mesangial cell proliferation,mesangial area widened and renal interstitial infiltration of inflammatory cells,etc) and electron microscopy revealed the presence of foot process integration and electron-dense deposit.Conclusion The mixture of paraphenylenediamine and H2O2 played a certain role on inducing lupus-like symptoms in mice. Objective To study the effect of the PPD-H2O2 mixture on the content of malondialdehyde(MDA) and glutamylcysteinylglycine(GSH) in liver of BALB/c mice.Methods Twenty-one female BALB/c mice were randomly divided into three groups of high PPD(n=7, 800mg/kg), low PPD(n=7, 400mg/kg) and control group(n=7, saline), cutaneous exposure.After 12 weeks of treatment, the mice were sacrificed and the livers was taken and homogenized.The content of protein and MDA was detected using Detect Kit,and the content of GSH was detected using Griffith.Results The content of MDA in liver tissue of the two experimental groups and control group showed significant differences (F=12.766, P<0.001).The content of MDA in liver tissue of the two experimental groups (high-PPD group and low-PPD group) were higher than that of control group (P<0.001), but there was no significant difference between the high-PPD group and the low-PPD group MDA content (P>0.05). The content of GSH in liver tissue of the two experimental groups and control group also showed significant differences (F=110.288, P<0.001). Compared to control group,the content of GSH in liver tissue of the two experimental groups were lower (P<0.001), and the content of GSH in high-PPD mice liver tissue were significantly lower than that of the low-PPD mice liver tissue (P<0.001). After 12 weeks,the concentration of ANA and anti-dsDNA antibody had no correlation with the content of MDA,but was negatively correlated with the content of GSH. Conclusions The mixture of paraphenylenediamine and hydrogen peroxide can reduced the ability of antioxidation in mice,and increased the level of lipid peroxidation.The level of autoantibody and lipid peroxidation/antioxidant capacity were closely related. |
