Objective To observe the funtion of the basic fibroblast growth factor(bFGF) in diabetic rats with acute myocardial infarction(AMI) in the area of myocardial infarction , cardiac function and the expression of the early gene heart transcription factor NKx2-5,GATA-4.Methods 27 GK rats (Type 2 diabetic rats) and 30 Wistar rats(nondiabetic rats) were involved in this study . AMI was induced in the rat model by irreversible left coronary ligation. The AMI rats were divided randomly into two groups.bFGF or PBS, were administered by intramyocardial injection into the ischemic ventricular wall. Sham-operated rats were given the PBS as the same way as bFGF.After two weeks,the rats were sacrificed and then were measured cardiac function by Langendorff perfusion device ,measured the myocardial infarction area by 0.5 % NBT , observated the expression of early heart transcription factor NKx2-5 and GATA-4 by Reverse transcriptase-polymerase chain reaction(RT-PCR).Results①Compared to the sham-operated group, all AMI rats showed significant left ventricular dysfunction; Compared to the PBS-treated group, Left ventricular systolic pressure (LVSP mmHg), left ventricular pressure change (+ LV dp / dt max mmHg / s) , and left ventricular ejection fraction (LVEF%) assessed in the Langendorff mode, were significantly improved in the bFGF--treated group,and left ventricular end-diastolic pressure (LVEDP, mmHg) were significantly decreased in the bFGF--treated group .Compared to the DM rats ,left ventricular function in the NDM rats were improved more significantly。②Infarcts area were significantly decreased in the bFGF--treated group, compared to the PBS-treated group.And in the NDM rats , the effection of bFGF was more obviously .③Compared to the sham-operated group and the PBS-treated group , the expression of myocardial development related genes NKx2-5 and GATA-4 in the bFGF-treated group were increased obviously. Compared to the DM rats ,the expression of NKx2-5 and GATA-4 in the bFGF-treated group in the NDM rats increased obviously. The results suggested that bFGF can promote early myocardial transcription factor expression and improve the cardiac function in the state of the high blood sugar. At the same time, bFGF may be adopted to stimulate the regeneration of myocardial cells, so as to achieve the improvement of cardiac structure and function. Conclusion The results suggested that bFGF improve the heart function of myocardial infarction in the T2DM rats and decrease the area of myocardial ischemia and promote the expression of early cardiac transcription factor NKx2-5 and GATA-4,but the effection of bFGF decreased in the state of high blood suger.At the same time, bFGF may be adopted to stimulate the regeneration of myocardial cells, so as to achieve the purpose of improving cardiac structure and function.
