| The model of subchronic aluminium poisoning chicken was estabolished by continuous peritoneal injection of fixed volume and different concentration gradient aluminium trichloride.The content of aluminium in blood and cerebellum, the percentage of cell apoptosis, DNA damage, the metabolism of nitricoxide and antioxygen function were detected by flame-atomic-absorption- spectrophotometer method, flow cytometry, spectrophotometry and DNA Ladder, and cerebrum and cerebellum were observed by microscope and electron microscope. The results were showed as follows.1 Aluminium poisoning model was induced successfully in the chicken by clinical symptom and dissection change.2 The concentration of Hb in group L, M and H was lower than that of in group C(p<0.01), which indicted aluminium resulted in chicken anemia.3 The histopathological changes of cerebrum and cerebellum were not significant in L group and control group. It was discovered that cerebrum centrum cells swelled, atrophied, necrotized, nucleolus pyknosis,vacuolar degeneration and quantity decrease of in cerebrum centrum cells and cerebellum Purkinje's cells in the M and the H group. The ultrastructural changes of cerebrum and cerebellum cell were not significant in the group L and C. Cerebrum and centrum cells swelled lightly, neurofilament and microtubule disorganized, interweaved and displayed vorticity apoptosis.Mitochondria swelled and emerged vacuolar, cristae broke, dissolved,even disappeared, and partial matrix dissolved and disappeared in the group H and M; In cerebellum Purkinje's cell,cytomembrane and karyotheca dissloved, cytochondriome cristae ruptured, dissloved,even disappeared.4 Cytomembrane shinking, budding, diluting matrix chromosome pyknosis, margin and disinter- grating the typical apoptosis characters were discovered in fluorescence microscope by AO/EB double fluorescent labeling.It showed that aluminium caused apoptosis in cerebrum and cerebellum.5 Apototic peak appeared before G0/G1 peak in aluminium poisoning chicken, proliferation index was lower and apoptosis rate was higher in the trial groups than those of in the control group (p<0.01). The results indicated that the proliferations of cerebrum and cerebellum were inhibited and cell population of apoptosis increased in chickens with aluminium expose.6 DNA Ladder analysis showed that DNA Ladder-shaped was displayed by analysis of DNA Ladder, which indicted that DNA was damaged in cerebrum and cerebellum in aluminium poisoning chickens.7 The content of aluminium of in aluminium poisoning groups was higher than that of the control group(p<0.05), and the contents of Ca, Fe, Cu, Zn and Mn in aluminium poisoning groups were lower than that of the control(p<0.01), which indicted that aluminium resulted in the disorder of microelement metabolism.8 The activity of nitric oxide synthase and the content of nitric oxide reduced, which indicted that NO message tranmitting access was blocked.9 Aluminium induced the decreased GSH-Px, SOD activity and the increased MDA content,which showed that aluminium induced oxidative damage of cerebellum and cerebrum in chickens. |
