Objective: TCCB and RCC are common tumor in urinary system. Their morbidity and mortality rate have rising tendency. The genesis, development, infiltration and metastasis of tumor are related to angiogenesis. VEGF has dominant position in angiogenesis factors. However,a new VEGF isoform which inhibits tumor growth thorough inhibiting angiogenesis were found in recent years. To further understand the role of VEGF165b in TCCB and RCC, differential expression of VEGF165b in TCCB and RCC and their normal tissues was investigated by immunohistochemistry and RT-PCR.Methods: The expression of VEGF165b protein was detected in 38 specimens of TCCB tissues, 36 specimens of normal bladder transitional cell tissues, 30 specimens of RCC tissues and 29 specimens of normal renal tissues by immunohistochemistry. The expression of VEGF165b mRNA was detected in 55 specimens of TCCB tissues, 43 specimens of normal bladder transitional cell tissues, 32 specimens of RCC tissues and 30 specimens of normal renal tissues by RT-PCR.Results: 1. The expression of VEGF165b in TCCB tissues and normal bladder tissues: (1) The positive rate of VEGF165b protein was 97.22% (35/36) in normal bladder tissues. The positive rate of VEGF165b protein was only 21.05% (8/38) in TCCB tissues. The differential expression of VEGF165b in the TCCB and normal bladder tissues had statistical significance (P<0.05). (2) The expression rate of VEGF165b mRNA in normal bladder tissues was 95.35% (41/43), while that of TCCB tissues was only 18.18% (10/55). The positive expression rate of VEGF165b mRNA in normal bladder tissues was significantly higher than that in TCCB tissues (P<0.05). (3) The expression of VEGF165b was related to clinical stage and pathological grade in TCCB tissues (P<0.05).2. The expression of VEGF165b in RCC tissues and normal renal tissues: (1) The positive rate of VEGF165b protein was 96.55% (28/29) in normal renal tissues. The positive rate of VEGF165b protein was 20.00% (6/30) in RCC tissues. The differential expression rate of VEGF165b in RCC and normal renal tissues had statistical significance (P<0.05). (2) The expression rate of VEGF165b mRNA in normal renal tissues was 93.33% (28/30), while that of RCC tissues was only 18.75% (6/32). The positive expression rate of VEGF165b mRNA in normal renal tissues was significantly higher than that in RCC tissues (P<0.05). (3) The expression of VEGF165b was unrelated to clinical stage in RCC tissues (Pï¹¥0.05). Conclusions: The low expression of VEGF165b might play a role in genesis and progression of TCCB and RCC. VEGF165b might be a new therapeutic target for TCCB and RCC.
