| Objective;To investigate the effect of copper needles retained in hemangioma on the expression of a-SMA and NF-kBp65 as well as histological and pathological changes.Methods:1.Between march 2006 to January 2009,12 cases with hemangioma were selected from the Department of Plastic and Cosmetic Surgery,Third Xiangya Hospital;Central South University,including 7 males and 5 females.All cases understood and agreed to their treatments,2.The copper needles with a blunt pointand a circularity end,which were made of industrial copper wire with diameter of 3mmwere sterilized before treatment.All cases were retained 13-42 copper needles in the local hemangioma underanesthesia for 5-7 days.3.All the samples were dyed in hematoxylin-eosine,then we observed the distribution changes of the endotheliocyte,smooth muscle cells and perithelial cells under the microscope of Nikon Eclipse 80i and measured the expression level of thea-SMA and NF-kBp65 by use of immunohistochemistry technique,then we took pictures by high resolution Nikon DS and NIS-Elements AR 3.0 software.Make semiquantitative analysis and figure collection by Ver.3.00 procedure software.4.The paired t test was applied to compare the difference of measurement data after treatment with that one before treatment.Results:All the 12 cases with hemangioma were involved in the result analysis.1.Histological and pathological changes:Before treatment.the lood vessels of neoplasm presented universally irregular and cystoid distention with varied diameter,however,tissue necrosis,thrombogenesis,ssociated with chromic inflammatory reaction,dwindling of blood essel and vessel wall thickening appeared in most specimens after copper needles. treatment.Fibrosis and granulomas could be seen in some specimens.2.The expression of a-SMA was significantly higher after treatment than before in hemangiomas(P<0.05)3.The expression of NF-kB p65 was significantly higher after treatment than before in hemangiomas(P<0.05)Conclusion:1.After copper needle retained,the lumen of blood vessels diminution and narrowed in hemangiomas and endotheliocytes and smooth muscle cells decreased obviously,however,the fibers surrounding the blood vessels grown in number.Some of the blood vessels atrophied and obliterated.2.After treatment,the expression levels of NF-kBp65 and a-SMA in hemangiomas lower than before treatment apparently. |
