Exhaustive Swimming Exercise On Rat Muscle Cells Muscle Actin With Actin And Its Mrna | | Posted on:2010-06-09 | Degree:Master | Type:Thesis | | Country:China | Candidate:H Wu | Full Text:PDF | | GTID:2204360275496390 | Subject:Human Movement Science | | Abstract/Summary: | PDF Full Text Request | | Objective The changes of actin and nebulin and their mRNA were observed in SM,CM and SMM after rat's single exhausted swimming and seven consecutive daily exhausted swimming.To explore the effects of exhausted swimming on muscles of rat,and provide morphological data for the further study of the exercises to the muscle cytoskeleto's injury.Method After exercises of SE and SCDE,rat's SM(quadriceps femoris and soleus muscle),CM(left atrium and right ventricle) and SMM(abdominal aorta and gastric wall) were cut off at 0H,1H,3H,6H,12H and 24H.These specimens were treated respectively with regular procedures of immunohistochemistry and in situ hybridization.The paraffin sections werestained by IHC(Envision Method) and ISH, the distribution were observed under the light microscope and calculate their changes of mean optics density and percentage of stained areas with the image analysis software.Result (1)SM:Actin and nebulin of QF and SOL continuoursly decreased from 0H to 3H after SE and their mRNA continuoursly increased compared with the exhausted swimming control.6H after SE,actin and nebulin began to increase and their mRNA were keep in a high level.12H to 24H after SE,actin and nebulin continuoursly increased to the level of EC,and their mRNA continuoursly decreased to the level of EC.The change of actin and nebulin of QF and SOL after SCDE were similar to the group of SE.(2)CM:Actin and nebulin of LA and RV continuoursly decreased from 0H to 6H after SE and their mRNA continuoursly increased compared with the EC.12H after SE,actin and nebulin began to increase and their mRNA were in a high level.24H after SE,actin and nebulin continuoursly increased and their mRNA continuoursly decreased,but both of them were not achieve the level of EC.Actin and nebulin of LA and RV continuoursly decreased to the lowest at 3H after the last swimming of SCDE and their mRNA continuoursly increased compared with the EC.6H after SE,actin and nebulin began to increase and their mRNA were keep in a high level.12H to 24H after the last swimming of SCDE,actin and nebulin continuoursly increased and their mRNA continuoursly decreased,but both of them were not achieve the level of EC.(3)SMM: Actin and nebulin of AO SMM and GW SMM were not changed obviously after SE compared with the EC.Actin and nebulin of AO SMM continuoursly decreased from 0H to 6H after the last swimming of SCDE and their mRNA continuoursly increased compared with the EC.12H after SCDE,actin and nebulin began to increase and their mRNA were in a high level.24H after SCDE,actin and nebulin continuoursly increased and their mRNA continuoursly decreased,but both of them were not achieve the level of EC.Actin and nebulin of GW SMM were not changed obviously after SCDE compared with the EC.Conclusion (1)Actin and nebulin have different expression in all of the SM,CM and SMM,they are the important component of the thin filament in cytoskeleto.(2)The exercises of SE and SCDE have different effects on the actin and nebulin in rat's three types of muscles.The effect of cardial muscle is great than the effect of skeletal muscle and smooth muscle.
