Objective: To investigate the expression and the clinical significance of survivin,c-myc and PCNA protain in esophageal squamous cell carcinoma.Methods: The expression of Survivin, C-myc and PCNA proteins were examined byusing immunohistochemical techniques(S-P) assay in 62 cases of esophagealsquamous cell carcinoma and 32 cases of esophageal dysplasia mucosa and 21 casesof normal esophageal mucosa. Laboratory data were then analysed statisticallytogether with the related clinical and pathological data.Results: Survivin was mainly expressed in cytoplasm and occassionaly expressed innucleus;C-myc was mainly expressed in nucleus and also could be found in cytoplasm.PCNA was mainly expressed in nucleus.1 Expressions of survivin in various lesions of esophageal epithelial :The expression of survivin in esophageal squamous cell carcinoma (69.4%) was significantly higher than that of in esophageal dysplasia mucosa (35.3%)and normal esophageal mucosa(0%),The positive expression in three groups of tissue were significant (x~2= 31.221, P=0.000).And the result showed that The expression of Survivin was related with the depth of invasion and metastasis of lymph node(P<0.05) .There was not statistic significance among varing degrees of differentiation .2 Expressions of C-myc in various lesions of esophageal epithelial:The expression of C-myc in esophageal squamous cell carcinoma(64.5%) was significantly higher than that in esophageal dysplasia mucosa (38.2%)and normal esophageal mucosa(9.5%), The positive expression in three groups of tissue were significant (x~2 = 18.978, P=0.000) .the expression of C-myc was related with the degrees of differentiation, the depth of invasion and metastasis of lymph node(P<0. 05).3 Expressions of PCNA in various lesions of esophageal epithelial:The expression of PCNA in esophageal squamous cell carcinoma(72.3%) was significantly higher than that in esophageal dysplasia mucosa (44.1%)and normal esophageal mucosa(14.3%), The positive expression in three groups of tissue were significant (x~2 = 21.860, P=0.000). the expression of PCNA was related with the degrees of differentiation, the depth of invasion and metastasis of lymph node(P<0.05).4 The relationship between survivin protein and C-myc protein:There was a close correlation between the expression of Survivin and C-myc protein in the esophageal carcinoma tissues (Spearman correlation coefficient r=0.531,P=0.000).5 The relationship between survivin protein and PCNA protein:There was a close correlation between the expression of Survivin and PCNA protein in the esophageal carcinoma tissues (Spearman correlation coefficient r = 0.376, P =0.003).6 The relationship between C-myc protein and PCNA protein:There was a close correlation between the expression of C-myc and PCNA protein in the esophageal carcinoma tissues (Spearman correlation coefficient r =0.300,P=0.018).Conclusion:1. The expression of Survivin,C-myc and PCNA in esophageal squamous cell carcinoma was significantly higher than that in esophageal dysplasia mucosa and normal esophageal tissues,which show they were highly associated with the occurrence and development of the esophageal squamous cell carcinoma.2. Survivin was associated with the depth of invasion and lymph node metastasis, which show that Survivin can be used as an important indicator of invasion and metastasis of the esophageal squamous cell carcinoma . It is expect to becom a new target of treatment for esophageal squamous cell carcinoma.3. The expressive intensity of C-myc and PCNA were closely correlated with histopathological grade, depth of invasion and lymph node metastasis. Both of them may be served as the molecular biological index of evaluating infilitration and metastasis.4. Positive correlation were found among Survivin, C-myc and PCNA which reveal that they may co-act in the development of esophageal squamous cell carcinoma.United detecting the expreesion of Survivin, C-myc and PCNA may be useful for diagnosis of early esophageal squamous cell carcinoma and evaluatingprognosis in esophageal squamous cell carcinoma.
