Abstract: In this dissertation, the phytochemical studies were carried out on Platycodon grandiflorum, and an accurate RP-HPLC method for the determination of eight saponins in Platycodon grandiflorum with three different detectors has been developed. In adition, the chemical and pharmacological research works about saponins in Platycodon grandiflorum were reviewed.Fifteen compounds were isolated from the root of Platycodon grandiflorum by various chromatographies. On the basis of chemical and spectral evidence, their structures were determined as follows: Platycodin D (J1); Polygalacin D2 (J2); Platycodin D3 (J3); Deapio-platycodin D3 (J4); Platycodin D2 (J5); Deapio-platycodin D2 (J6); Platycoside E(J7); Deapioplatycodine E(J8); Platycogenin A(J9); Platycogemin B (J10); Tangshenoside II(J11); Seringin (J12) and Mannitol (J13). Compounds J9 and J10 were new compounds. Compounds J11, J12 and J13 were obtained from Platycodon grandiflorum for the first time.An accurate RP-HPLC method for the determination of eight saponins (J1-J8) in Platycodon grandiflorum with three different detectors (HPLC-ELSD, HPLC-DAD and HPLC-NQAD) has been developed. This reseach work will provide scientific evidence for the quality control as well as the evaluation method for breeding improved varieties of Platycodon grandiflorum.In this paper, the structural characteristics and spectroscopic features and separation method for triterpenoid saponins from Platycodon grandiflorum were summarized. In addition, the analysis methods of three different detectors were discussed.
