Objective: To investigate hepatic energe metabolism and the effect of isoflurane and sevoflurane preconditioning on hepatic ischemia-reperfusion injury in rats.Method: Sixty adult male SD rats weighing 240250g were randomly assigned into 3 groups (n=20),each:Control group(IR), no volatile anesthetics ; Sevoflurane group(Sev), 2.0% sevoflurane; Isoflurane group(Iso),1.5% isoflurane. Thirty minutes after the start of volatile anesthetics(except group IR)(T1), then rats were subjected to 30 min hepatic ischemia and 30 min(T2) and 120 min(T3)of reperfusion. Rats were killed at the end of ischemia, serum levels of tumor necrosis factor-α, IL-1β, alanine aminotransferase (ALT), aspartate aminotransferase (AST) lactate dehydrogenase (LDH) content were test in T1, T2, T3; We also measured liver tissue glutathione reductase(GR), malondialde(MDA) content, superoxide dismutase(SOD) activity and hepatic ATP in T3.And part the liver pathology did HE staining . Results: Compared to each group's T1, the level of ALT, AST, LDH in serum of rats increase significantly in each group's T2 and T3 (p<0.05). At T1, the level of ALT, AST, LDH in the serum have no significances difference in these 3 groups (p>0.05). At T2 and T3, compare to group IR, the level of ALT, AST, LDH in serum of rats decreased significantly in group Sev and group Iso (p<0.05). In these 3 groups, compare to T1,the level of TNF-α,IL-1βin serum increased significantly in T2 and T3(p<0.05). At T2 and T3, compare to group IR, the level of IL-1βin serum decreased significantly in group Sev (p<0.05), while there are no difference in group Iso. At T2 and T3, in these 3 groups, there are no differences in TNF-αin scrum (p>0.05). Compare to group IR, the level of MDA in hepatic tissue decreased significance in group Sev and group Iso (p<0.05), while the level of SOD, ATP and GR in hepatic tissue increased significance (p<0.05). Compare to normal hepatic tissue, there groups'liver pathology show the change of ischemia. But the difference among these 3 groups is not significance.Conclusion: Administration of isoflurane and sevoflurane before ischemia can reduce the damage of ischemia reperfusion in the liver. It can also improve the hepatic energy metabolism of the liver suffered from ischemia-reperfusion injury.
