Background and Objective:As a newly discovered member of B7 family, costimulatory molecule B7-H3 expressed on various of immune cells such as antigen presenting cell. With the research found that B7-H3 molecule also could express abnormally in various of tumors cells that may be associated with tumor progression. Therefore, B7-H3 could be expected to become a new targeted marker for tumor diagnosis, therapy and prognosis. To study expression of B7 family's new member B7-H3 in breast cancer and clinical significance.Methods:B7-H3 mRNA expression were examined using a real-time fluorescence quantitative reverse transcription polymerase chain reaction (PCR) in paired breast tissue samples from carcinoma tissues and adjacent tissues. B7-H3 protein expression was examined using immunohistochemistry in breast tissues and normal tissues samples from patients with breast cancer. Relationship between B7-H3 mRNA expression and protein expression and various clinicopathologic variables were analyzed.Results:(1) B7-H3 mRNA was expressed in both carcinoma tissues and adjacent tissues, but it was significantly higher in breast carcinoma tissues than in adjacent tissues (z=-2.582, P<0.05)(2) There was no statistic difference between B7-H3 mRNA expression and clinicopathologic variables (P> 0.05).(3) The positive rate of B7-H3 protein was 68.75%(22/32) in breast cancer, and there was no expression in normal tissues.(4) B7-H3 protein expression was positively correlated with patient's tumor size and pathological stage (P< 0.05)Conclusions:The higher expression of B7-H3 existed in breast carcinoma tissues, and B7-H3 protein expression was significantly correlated with tumor size and pathological stage.
