ObjectiveTo investigate the consistency of Her-2 protein by immunohistochemistry(IHC) and Her-2 gene amplification by fluorescence in situ hybridization(FISH)on the basis of standardized testing in invasive breast cancer and discussed the clinical pathological significance of test results in breast cancer, researched the impact of Polysomy 17 on Her-2 testing further.MethodsCollected 96 cases of paraffin embedding specimens which invasive breast cancer patients without preoperative treatment on the requirements of standardized, The expression of Her-2 protein and Her-2 gene amplification in breast cancer were detected by immunohistochemical staining and FISH, Analyzed the consistency between the two methods respectively,And the results were demonstrated by either absolute Her-2 gene copy number the ratio chromosome 17. ResultsThe results coincidence rate of FISH and IHC was 88.9%(k=0.739 P<0.05).in 96 cases ofinvasive breast cancer,IHC(+++)in 11 cases(11/96),IHC(++) in 14 cases(14/96), IHC( ±) in 71 cases(71/96),FISH detectd the results for samples IHC(+++), her-2 gene amplification was 90.9 %(10 cases were positive), The FISH coincidence rate of IHC(++) specimens was 71.4 %( 10 cases were positive), IHC( ±) and FISH detection coincidence rate 95.3%( 4 cases were positive), the difference among three groups was statistically significant(p < 0.05), the results of FISH detect suspicious cases more than 17 dyeing effect. Conclusions(1) IHC(+++) / IHC(±) samples with FISH detection results have good consistency, The difference between IHC(++) samples and FISH detection was larger.So the IHC can be used for early screening her-2 protein expression method.(2)FISH can be used as the IHC breast cancer(++) samples of the effective method in her-2 gene diagnosis 17 polysomy affect the test results of Her-2, and it is main reason for generating suspicious results in Her-2 testing.
