| Objective:Through the detection of Fragile histidine triad (FHIT) and Ki67 antigen in normal cervix tissues,cervical intraepithelial neoplasia (CIN) and cervical squamous cell cacinomas(SCC),we observe their expression in different cervix lesions,to investigate the change trend and the mutual relationship of FHIT protein and Ki67 antigen from nomal cervix tissue-precancerous lesion-cancer process.Methods:Immunohistochemical analysis was used to detecet the expression of FHIT and Ki67 antigen in normal cervix tissues,CINâ… ,CINâ…¡,CINâ…¢and cevical squamous cell carcinoma of the 10 cases,16,18,18,17 cases each,which had been histologically confirmed and were divided into control group and experimental group.Results:1. The negative rate of FHIT expression was found in 0.00%(0/10) of the normal cervix tissues,6.25%(1/16) of the CIN I grade,27.78%(5/18) of the CIN II grade,22.22% (4/18) of the CIN III grade,64.71%(11/17) of the cevical squamous cell carcinoma. Each group of FHIT protein expression differences(P<0.01). With the rising grade of lesions, the expression level of FHIT protein descend(P<0.005, C=0.9104).2. The positive rate of Ki67 expression was found in 10.00%(1/10) of the normal cervix tissues,56.25%(10/16) of the CIN I grade,72.22%(13/18) of the CINâ…¡grade,83.33%(15/18) of the CINâ…¢grade and 88.24% (15/17) of the cevical squamous cell carcinoma. There was a significant difference in the expression of Ki67 antigen between the groups (P<0.05). With the rising grade of lesions, the expression level of Ki67 antigen showed rising trend(P<0.01, C=0.3440).3. There was a negative correlation between FHIT and Ki67(r=-0.760, P<0.05).Conclusion:1. With cervix lesions from normal tissue to cervical cancer development process, FHIT protein expression is gradually descend, however, Ki67 antigen expression is gradually ascend.2.The negative expression of FHIT protein and the positive expression of Ki67 antigen can be used as a high degree of clinical prediction of pre-cancerous lesion to invasive carcinoma of useful biomarkers.3. Detection of FHIT protein and Ki67 antigen can be used to guide the clinical treatment of CIN,so it can be avoided the excessive and inadequate treatment. |
