Fhit, Bax And Ki67, And Cervical Cancer Relationship And Chemotherapy Drugs On The Expression Of Impact Study | | Posted on:2010-07-08 | Degree:Master | Type:Thesis | | Country:China | Candidate:Z P Shi | Full Text:PDF | | GTID:2204360275461476 | Subject:Obstetrics and gynecology | | Abstract/Summary: | PDF Full Text Request | | Objective:(1)To investigate the inhibitory effects of cisplatin and Interferon-γon cervical carcinoma cell Hela in vitro.(2)To sudy the expression of FHIT,Bax and Ki67 induced by cisplatin and Interferon-γon cervical carcinoma cell Hela in vitro.(3)To sudy the expression of FHIT, Bax and Ki67 in cervical carcinoma.To evaluate the correlation between them and clinico-pathologic variables in these cervical carcinoma patients.Methods:(1)The effect of cell cycle and cell apoptotic rates were analyzed by MTT assay and flow cytometry(FCM).(2)The expression of FHIT, Bax and Ki67 were analyzed by immunocytochemistry.(3)The expression of FHIT, Bax and Ki67 were examined by immunohistochemistry method and tissue chip method in 60 cases of cervical carcinoma tissue.Results:(1)By MTT assay,Cisplatin,Interferon-γand both inhibited proliferation in a time and dose-dependant manner(P<0.05),accelerated the apoptosis of Hela cell in a dose-dependant manner(P<0.05).(2)By immunocytochemistry,when incubated with Cisplatin,Interferon-γand both, it was found that the negative expression of FHIT and Bax were increased and the the positive expression rates of Ki67 were statistically declined in vitro.(3) Reductionor absence of FHIT expression was found in 58% (29/50) of the tumors.Positive staining of Ki67 was detected in 43 of 50 (86%) cervix carcinomas.Bax immunore activity was observed in 31 of 50 (62%) cervix carcinomas.Loss of FHIT was irrelevant with histological grade and lymph nodes metastasis,as well as age,clinical stages and histological type.The expression of Ki67 correlated significantly with histological grade and clinical stages.Loss of bax increased with histological grade.There was a positive correlation between FHIT and bax, and a negative correlation between FHITand Ki67.Conclusions:(1) Both cisplatin and Interferon-γinhibited the proliferation and induced the apoptosis of Hela cell lines in vitro.(2) By immunocytochemistry,when incubated with cisplatin and Interferon-γ,it was found that the expression rates of FHIT and Bax were increased and the positive expression rates of Ki67 were statistically declined.(3)The unexpression of FHIT,Bax and the overexpression of Ki67 may play important roles in carcinogenesis and development of cervical carcinoma,they were valuable to further understand the biolodigalbehavior of cervical carcinoma. | | Keywords/Search Tags: | Cisplatin, Interferon-γ, cervical carcinoma, Fhit, Bax, Ki67 | PDF Full Text Request | Related items |
