| Perennial Chinese herb Corydalis yanhusuo W. T. Wang(Papaveraceae) is a valuable drug in Changbai Mountains. Because of its unique pharmacological activities such as blood stasis, sedation and hypnosis, inhibition of gastric acid secretion, scholars have been interesting about the plant.In recent years, the chemical composition and biological activity of Corydalis yanhusuo were studies, some research results have been used in clinical. Pharmacological research has made some progress, but reports of Corydalis novel compounds, and the bioactive ingredients of Corydalis has rarely been reported, on the DPPH free radicals scavenging activity and tyrosinase inhibitory activity is virtually no literature. For the active ingredient of Corydalis, active ingredients with DPPH free radicals scavenging activity and tyrosinase inhibitory activity were studied.In this study, useing DPPH free radicals scavenging activity and tyrosinase inhibitory activity tests as a guide, the extracts of Corydalis yanhusuo were isolated. By means of macroporous resin column, silica gel column chromatography, ODS reversed phase column chromatography and HPLC ten compounds wrer purified from Corydalis. The stuctures of compound 1 and 2 were established as d-corydaline and tetrahydropalmatine, respectively, on the basis of MS, 1D and 2D NMR.In addition, the results of biological activity test of Corydalis extracts including DPPH free radicals scavenging activity and tyrosinase inhibitory activity showed that the inhibition rates of d-corydaline and tetrahydropalmatine on the DPPH were 95% and 61%, respectively, tyrosinase inhibition rates were 39% and 46% respectively. In terms of the overall composition, most of the components of the DPPH inhibition rates reached 90% or more, while some components showed activities both on tyrosinase and DPPH inhibition that the inhibition rates reached 90%. |
