Traditional Chinese Medicine Acanthopanax senticosus (Rupr.et Maxim) Harms. belongs to family Araliaceae. As Chinese medicine, the dry roots rhizomes, stems, leaves and pulp of this plant have been widely applied. This herb has been widely used for the treatment of Parkinson's disease, neurasthenia, diabetes mellitus, hypertension, arteriosclerosis and ischemic heart disease. According to the references, many compounds were isolated and identified as so far, such as steroid, triterpenoid saponin and so on.Six compounds were isolated from Acanthopanax senticosus (Rupr.et Maxim) Harms. by using ultrasonic assisted solvent extraction liquid-liquid extraction, normal and reverse phase column chromatography, recrystallization and semipreparative HPLC technology,. For the purposes of active compounds, bioassay-guided isolation method including DPPH radical scavenging activity test and tyrosinase inhibition activity test was used to guide the separation and purification of the extracts of this plant. The stuctures of compound 1 and 2 were established as syringaresinol and secoisolariciresinol, respectively, on the basis of MS, 1D and 2D NMR experiments, and secoisolariciresinol is reported for the first time from A. senticosus.The results of radical scavenging activity test and tyrosinase inhibition activity test for the different fractions of the extracts showed that the extracts and fractions inhibited the DPPH radical and tyrosinase. The inhibition ratios of fractions F7.4.3.2.2.1-F7.4.3.2.2.5 against DPPH radical were over 90%, while fraction F52 against tyrosinase was 50%. Compound 1-6 showed remarkable scavenging activity toward DPPH radical with IC50 values of 0.47,0.01,14.48,5.11,4.05 and 20.56μg/mL, respectively, while the positive control vitamin C with IC50 value of 1.46μg/mL. Compound 1-6 also showed inhibitory activity toward tyrosinase with inhibition ratios of 39%,35%,30%,31%,32%and 25%respectively at the concertration of 500μg/mL. To our knowledge, this is the first report of constituents from A. senticosus showed inhibition activity toward tyrosinase.
