| BackgroundColorectal carcinoma is one of the most common malignant tumors all over the world. It is hazardous to health of human beings. Metastasis is an important biological behavior of colorectal carcinoma. One of the major causes of lower survival rate is also metastasis. There are key significances for judge prognosis,instruct treatment, and improve quality of life to ensure metastasis potency. and its related factors of colorectal carcinoma.Metastasis involves various genetic alteration,complex pathological mechanisms, and multi-step processes, including metastasis-associated genetic mutation and activation, cancer cell adhesion,motility, proliferation,degradation of extracellulax matrix, tissue immunity,tunlor angiogenesis,ete. Then by gent expression regulation mechanisms, functional proteins regulate occurrence and development of tumor metastasis processes.The annexin superfamily consists of 13 calcium or calcium and phospholipid binding proteins with a significant degree of biological and structural homology (40-60%). First described in the late 1970s and subsequently referred to as macrocortin,renocortin, lipomodulin, lipocortin-1, and more recently Annexin1, this 37 kDa calcium and phospholipid binding protein is a strong inhibitor of glucocorticoid-induced eicosanoid synthesis and PLA2. Recent interest in the biological activity of this intriguing molecule has unraveled important functional attributes of Annexin 1 in a variety of inflammatory pathways, on cell proliferation machinery, in the regulation of cell death signaling, in phagocytic clearance of apoptosing cells, and most importantly in the process of carcinogenesis and the metastasis of carcinoma.ObjectionThis study was designed to use immunohistochemistry, fluorescence quantitative RT-PCR, and Westernblot to detected the expression levelof Anx A1 in colorectal cancer, Tubulovillous adenoma, the normal tissue and adenocarcinoma lymph node metastasis。Investigate the relationship between the expression level of Anx A1 and the occurrence and metastasis in colorectal cancer. Materials and MethodsCollection of 50 cases of colorectal cancer and 10 cases of tubulovill adenoma, who were from the Second Affiliated Hospital of Second Military Medicine University(2008-1010), use immunohistochemistry, fluorescence quantitative RT-PCR, and western blot to detected the expression level of Anx A1 in colorectal cancer, tubulovillous adenoma, normal tissue and adenocarcinoma lymph node metastasis.Collection more 20 primary patients with colorectal resected cancer and 10 patients with colorectal resected tubulovillous adenoma,use fluorescent quantitative RT-PCR and western blot to detect the expression level of mRNA and protein of Anx A1 in coIorectal.The use of SPSS statistical software for data processing and statistical analysis.Results1. Expression and signification of Anx A1 protein were detected in normal colorectal tissues,tubulovillous adenoma,colorectal carcinomas and lymph node metastasis of colorectal carcinomas by immunohistochemical staining and westernblotting.At the protein level, the expression of Anx A1 was low expression in normal group and up-regulation in adenocarcinoma and adenoma,and the difference was significant (H=16.913, P=0.000<0.05).In lymph node metastasis group,the expression of Anx Al in lymph node metastasis was stronger than the primary tumor,the difference was significant (H=4.79,0.025 0.05). Conclusion1. Anx A1 low expression in normal group and up-regulated in adenoma group and carcinomas group, closely related to tumor differentiation grades and positively related to lymph node metastasis of colorectal carcinoma;2. Anx A1 mRNA widely expressed in the intestinal mucosal, down-rugulated in carcinomas group, as well as closely related to tumor differentiation grades;3. Annexin I protein is an important marker for the biological behaviors and prognosis of colorectal carcinoma. |
