| Objective:In recent years, we have reseached the pathogenesis of type II diabetes thoroughly.It is a complex disease of the polygenic inheritance,which is formed for the genetic and environmental factors.Current research mainly focus on the insulin resistance andβcells secrete defect that is two main phase of the incidence mechanism of type II diabetes.The research seldom involves in the intestinal micro-ecology system because of the complexity of itself.Furthermore,it is more uncommon to prevent and cure type II diabetes by use of the lactobacillus.In the experiment,we observe for the disease process of the susceptible and illed mice,and the intestinal flora's change,sequently discuss their relations,which can provide experimental basis for the prevention and treatment of typeⅡdiabetes.Methods:Diabetic model was induced by injecting with streptozotocin (STZ).Mice were divided into six groups as normal control group (CON group),case-control group (DM group),Low-dose treatment group (L group). Medium-dose treatment group (M group),High-dose treatment group (H group),Prevention group (Y组).IF one mouse's fasting blood glucose(FPG)≥11.1mmol/L while it was absolute diet 12 hours before,the mouse will be recognized as a diabetic model.FPG,weight,C-Peptide,volume of blood-fat were detected on the day before STZ injected,one day,12 days,25 days after the modeling.25 days after the modeling and mice were anatomized, pancreas treaded by HE staining and observed the situation of intestinal flora.Results: 1. After intraperitoneal injection of STZ, Y group incidence rate is 80%, significantly lower than other groups (100%), time of onset was signify-cantly delayed and significantly reduced the degree of islet inflammation.2.When FPG is increasing, DM group, L group, D group mice lost their weight significantly, H group, Y group mice had to reduce weight. C peptide levels are lower than normal. Reduce the number of probiotics. After taking Lb.casei agents in mice, reducing blood glucose, body weight returned to normal, C peptide levels increase and the number of probiotics has been resumed. However, certain conditions pathogenic bacteria such as E. coli, staphylococcus was increased throughout the experiment in the state.3.STZ induced type II diabetes, the levels of TG, LDL were increase, while the levels of TC, HDL did not change significantly.25 days after administration, mice in each group elevated TG, LDL dropped in varying degrees, Y mice returned to normal levels.4.Pathological structural changes in mouse pancreatic:Light micros-cope, CON mice perimeter between areas of Langerhans and exocrine gland is clearl, areas of Langerhans surface coated with a layer of fibrous membrane, the shape, neatly arranged in the pancreatic islet cells, the same size distribution rules, the nuclei were stained blue, big, round, clear nucleus, prominent nucleoli, abundant cytoplasm was stained pale pink. DM Group islet atrophy, as the proliferation of pancreatic islet cell division, or hyaline degeneration, B cells were decreased, cell sizes, pancreatic islet cell hyperplasia, the side of the majority is small blood vessels, apoptotic cells. L, M, H group of pancreatic islet cell number and on the island more than the DM group, form a more rules, a small number of nuclear condensation, vacuolar degeneration of some cells. Y group pancreatic islet cell is like normal, cells more regularly arranged, pancreatic islet number and cells than L, M, H group, cell distribution uniformity, nuclear size basic equal small karyopyknosis.Conclusion:The Lactobacillus acidophilus on STZ induced type II diabetic mice protecte structure and function of injury protection. Type II diabetes STZ-induce mice establish intestinal Bifidobacterium and Lactobacillus trend of negative correlation with blood glucose. while Enterobacter, Staphylococcus and the variation of blood glucose is not obvious. |
