Background/Aim:With the change of modern people’s dietary structure,the increase of work rhythm and the increase of mental and social pressure,constipation has become one of the important factors that haunt people’s life,which greatly reduces people’s quality of life.Constipation will not only cause the decline of intestinal peristalsis function caused by excretion difficulties and other common symptoms,but also accompanied by intestinal flora disorders,long-term constipation can even become an inducement factor for various diseases.In addition to improving the diet structure,increasing the amount of water to drink,exercise and other basic treatment,clinical often take laxatives,gastric motility drugs as the main means of treatment,but most constipation treatment drugs in the improvement of constipation symptoms at the same time there are a variety of potential side effects,Therefore,it is necessary to seek a moderate,safe and effective means of intervention and treatment.A large number of literature studies have found that single-strain probiotic preparations can improve the symptoms of constipation,increase the propulsion of intestinal contents,regulate the structure of intestinal flora and promote the excretion of faeces in vitro through animal constipation model experiments and clinical human intervention experiments.However,it is not clear whether the combined preparation of multiple strains can improve the symptoms of constipation and regulate the structure of intestinal flora.Therefore,the purpose of this study is to establish a compound diphenol ester induced constipation Model,the use of different doses of Lactobacillus acidophilus ncfm-bi-07-rat Lactobacillus NH001 triple probiotic preparation to the constipation mice to explore the effect of the new triple probiotics on constipation,At the same time,using the developed mature DNA gene sequencing 16 s r DNA method instead of the traditional petri dish bacterial culture counting method,intuitively and comprehensively explore whether the use of the new triple probiotic preparation can improve the intestinal flora structure of mice.Research methods:The experiment of Intestinal motion rate and the number of defecation granules were randomly divided into blank control group,model control group and three dose group with low medium and high height after 5 days of adaptive feeding in 50 km mice,and 10 in each group.The blank control group and the model control group were given distilled water,and the low middle and high dose group was given daily 0.165G/KG·BW,0.33G/KG·BW and 0.99G/KG·BW,respectively,and 24 hours after filling the stomach for 14 days,the model control group and the mice in each dosage group were 50mg/kg· Bw Oral compound diphenol ester suspension(i.e.0.1ML/10G·BW)established a mouse constipation model,the blank control group with the same amount of stomach to give distilled water,after 45 minutes and 5 hours after the observation of the small intestine ink propulsion rate in each group of mice,The number of defecation granules and fecal water content in each group of mice were calculated,and the time of first black defecation was recorded in each group of mice.In the experiment of intestinal flora regulation,10 were randomly selected in 42 BALB/c mice as their own contrast blank group,and after 5 days of adaptive feeding,all mice were randomly divided into blank control group and three dose group with low medium and high height,and the blank control group was given distilled water,and the dosage group was given the same dose to the triple probiotic preparation for gastric lavage.Self-comparison of the blank group in the stomach before the aseptic collection of mouse rectal faeces,all mice filled the stomach 30 days after the Aseptic collection of rectal faeces,the use of 16 s r DNA gene sequencing of the flora DNA diversity and the determination of various horizontal flora species.Research results:1)In the experiment of Intestinal motion rate,the experiment of defecation granules and the regulation of intestinal flora,the gastric group was fed by distilled water,low middle and senior dosage group for 14 days and 30 days without any effect on its weight,the hair color was normal and the mental state was good.The results showed that the short and long term use of different doses of the new triple probiotics had no effect on the weight gain of mice,and there was no adverse reaction to the growth of mice.2)In the Intestinal motion rate experiment,the Intestinal motion rate in the Model control group was significantly lower than that in the blank control group(P<0.05),suggesting that the molding was successful.After statistical analysis,the value of ink propulsion rate in mice with low,medium and high dose groups was higher than that in the model control group,and the propulsion rate of small intestine in mice with high dose group was significantly higher than that in the Model control group(p<0.05).3)In the experiment of defecation particle number,the first black time of the model control group and the number of 5-hour defecation particles were significantly lower than that of the Blank control group(P<0.05),which suggested that the molding was successful.After statistical analysis,the first black defecation time and 5-hour defecation granules of mice in Low,medium and high dose group were significantly different from those in the Model control group(P<0.05),among which there were significant differences between the two indexes in the middle dose group and the control group(p<0.01).There was no difference in fecal water content index in each group of mice.4)In the experiment of intestinal flora Regulation,the difference index value of the Chao1 index,observed species index,Whole tree Index,Shannon Index and Simpson index used to estimate microbial community was significantly higher than that of the Blank control group(p< 0.05),it is explained that eating contributes to the formation of the diversity of intestinal flora in mice,and the diversity index of each dose group is significantly higher than that of the blank control group,which shows that giving the triple probiotic preparation is more helpful to the formation of intestinal flora in mice than to giving distilled water alone.At the same time,the number of probiotics,such as Bifidobacterium and Lactobacillus in the faeces of each dose group,increased significantly(P<0.05),the genus Escherichia coli,Clostridium,Enterococcus and other harmful bacteria decreased significantly(P<0.05).Conclusions:The new Lactobacillus acidophilus ncfm-Lactobacillus bi-07-Rat Lactobacillus NH001 can improve the symptoms of constipation in different degrees,and has a certain regulating effect on the structure of intestinal flora. |
