| Objective:To compare the expressing level of virulence factor GelE of clinical Enterococcus faecalis,and investigate the relationship between virulence factor GelE of Enterococcus faecalis and the symptoms of the clinic patients with failed endodontic treatment.Methods:The cases of Failed endodontic treatment including 151 teeth of 131 patients were collected(one single root canal was sampled each tooth),and recorded the clinical symptoms, signs and related information. Disinfencted the teeth,and removed the preexisting filling with sterile dental drill.Sterile files were used for the removal of contents of the root canal.The paper points were introduced to a level approximately 2-3mm short of the tooth apex based on the diagnostic radiographs,and retained for 60s to sample the bacteria. Extracted DNA from the clinical samples,and investigated the occurrence of Enterococcus faecalis by means of the polymerase chain reaction. Extracted total RNA from the clinical samples,and obtained cDNA by Reverse Transcription PCR, Virulence factor gelE was detected by Real Time Quantitative PCR, investigated the relationship between virulence factor gelE of Enterococcus faecalis and the symptoms of the clinic patients with failed endodotic treatment,which were detected consisted with Enterococcus faecalis.Results:1. 58 cases were detected in 151 root retreatment cases by means of the Polymerase chain reaction.The detection rate of Enterococcus faecalis is 38.4%. 2. Real Time Quantitative PCR was used for detecting the expression of Enterococcus faecalis virolence factor gelE in 58 root canal retreatment cases.57 cases were detected which had expressed Enterococcus faecalis virulence factor gelE. The expression rate of Enterococcus faecalis virulence factor gelE is 38.4%.49 cases were detected over-expressed;8 cases decreased;1 case no expression.3. The expression of Enterococcus faecalis virulence factor gelE in cases of root canal retreatment had closed correlation with periapical destruction and the clinical symptoms or physical signs by statistical analysis (P<0.05). The expression of Enterococcus faecalis virulence factor gelE in cases had root tip manifest aneretic and the clinical symptoms or physical signs increased.Conclusions:1. The expression of Enterococcus faecalis virulence factor gelE in cases of root canal retreatment had closed correlation with the clinical symptoms or physical signs. The expression of Enterococcus faecalis virulence factor gelE in cases had clinical symptoms or physical signs increased.2. The expression of Enterococcus faecalis virulence factor gelE in cases of root canal retreatment had closed correlation with periapical destruction. The expression of Enterococcus faecalis virulence factor gelE in cases had periapical destruction increased. |
