| Objective:To study the effect of doxycycline and allicin reference-dially trisulfide (DATS) on the genetic expression and virulence factors of gelE and esp, which were related in the formation of Enterococcus faecalis biofilm. Also to provide some experimental evidence, for the use of allicin as a root canal medication.Methods:MTT assay was used to describe the growth curve of Enterococcus faecalis and to establish the Enterococcus faccalis biofilm model of exponential phase, stable phase and starvation phase. Different concentrations of DATS;1280μg/ml,2560μg/ml,5120μg/ml, and doxycycline;80μg/ml,160μg/ml,320μg/ml, were applied to the model of each phase. The genetic expression of virulence factors gelE and Esp in each model with the method of real-time PCR was determined.Result:1. The genetic expression of gelE and esp before medication is less than that after medication, the difference is statistically significant (p<0.05).2. The genetic expression of esp decreased with the increase in the concentration of DATS or doxycycline, the difference is statistically significant (p<0.05). At exponential phase, the difference of gene esp expression between the group of2560μg/ml,5120μg/ml DATS and the group of160μg/ml doxycycline is not statistically significant (p>0.05). At stable phase and starvation phase, the difference of gene esp expression in the group of2560μg/ml,512μg/ml DATS and in the group of160μg/ml doxycycline is not statistically significant (p>0.05).3.The genetic expression of gelE decreased with the increase of the concentration of DATS or doxycycline, the difference is statistically significant (p<0.05). At exponential phase and stable phase, the difference of gene gelE expression between the group of5120μg/ml DATS and the group of160μg/ml doxycycline is not statistically significant (p>0.05).4. The genetic expression of gelE and esp at starvation phase is more than that in stable phase or in exponential phase, the difference is statistically significant (p<0.05).Conclusion:1. DATS and doxycycline has the ability to inhibit the genetic expression of virulence factors gelE and esp.2. The inbibitional effect of DATS with higher concentration (5120μg/ml,2560μg/ml) is equivalent to that of doxycycline with lower concentration (160μg/ml,80μg/ml) on the genetic expression of gelE and esp. 3. The genetic expression of gelE and esp at starvation phase is more than that at exponential phase and stable phase. |
