Objective:To estimate the expressions of FHIT, PTEN and Ki-67 in the adult-type granulosa cell tumors of the ovary(AGCTO) and analyze the relationship between their expressions and clinical phathological parameters.The aim of the study is for better and clearer understanding of its mechanism and to provide reliable joint-detection indicators for screening the high-risk groups and early diagnosis on molecular level.Methods:The expressions of FHIT, PTEN and Ki-67 protein were detected in 45 cases of AGCTO by immunohistochemical method and the correlation between their expressions and the clinical staging, histological grade and lymph node metastasis was studied. Analyses were performed on SPSS 11.5.Results:l.The positive expression rate of FHIT is 66.7%in AGCTO, it was lower than that in normal ovarian tissues (100.0%)(P<0.05). The expression level in AGCTO patients with stage II+III was significantly lower than that in stage I (P<0.05), and lower in the poorly differentiated patients than in the better differentiated patients (P<0.05), and also lower in patients with postive lymph node metastasis than in patients with negative lymph node metastasis(P<0.05).2.The positive expression rate of PTEN is 64.4%in AGCTO, it was lower than that in normal ovarian tissues (100.0%)(P<0.05). The expression level in AGCTO patients with stage II+III was significantly lower than that in stage I (P<0.05), and lower in the poorly differentiated patients than in the better differentiated patients (P<0.05), and also lower in patients with postive lymph node metastasis than in patients with negative lymph node metastasis(P<0.05).3. Ki-67PI is higher in AGCTO group than in control group(P<0.05). The expression level in AGCTO patients with stageⅡ,Ⅲwas significantly higher than that in stageⅠ(P<0.05), and higher in the poorly differentiated patients than in the better differentiated patients (P<0.05), and also higher in patients with postive lymph node metastasis than in patients with negative lymph node metastasis(P<0.05). 4. There is negative correlation between the expression of FHIT, PTEN and Ki-67. There is association between the expression of FHIT and PTEN. The statistical evaluation shows that the positive expression rates of FHIT, PTEN and Ki-67 have close relation with clinical stages histological grade as well as lymph node metastasis.Conclusions:FHIT, PTEN and Ki-67 can be served as subsidiary markers of the tumors, And their expressions can provide a theoretical basis for predicting biological behaviors of AGCTO.
