Objective:Studing the expression of VEGF-D,VEGFR-3 in breast infiltrating cancer and the benign. Its relationship with lymphatic metastasis,grade and prognosis of breast cancer may be reported.Methods:Paraffin-embedded specimen from 20 cases of benign tumor and 60 cases of primary infiltrating ductal carcinoma were studied. VEGF-D,VEGFR-3 expression between benign and malignant breast tumor (lymph node metastasis group and no lymph node metastasis group) as well as the clinicopathological characterization were analyzed statistically.Result:In 60 case of breast infiltrating ductal carcinoma, the expression of VEGF-D,VEGFR-3 was difference.1,In 35 cases lymph node metastasis group, VEGF-D positive was 80.0%(the positive cases of infiltrating ductal carcinoma were 28). In 25 cases no lymph node metastasis group, VEGF-D positive rate was 36.0%(the positive cases were 9); respectively; the positive rate of VEGF-D was significantly higher in lymph node metastasis group than in no lymph node metastasis group(P<0.01).2,The positive rate of VEGFR-3,85.7%(30/35), in lymph node metastasis group, was significantly higher than that,40%(10/25), in no lymph node metastasis group(P<0.01).3,The VEGFR-3 positive rate was 81.1%in the 37 cases of VEGF-D positive cases(30/37), and significant correlation was found between the expression of VEGF-D and VEGFR-3 (P<0.05).4,The expression of VEGF-D was significant correlation between the lymph node metastasis and Cerb-2; no significant correlation with age,the tumor size,clinic grade,ER and PR(P>0.05).5,In 20 cases breast benign, VEGF-D positive rate was 35%(7/20); VEGFR-3 was 25%(5/20); and significant correlation was found between the expression in the benign and malignant(P<0.01).Conclusion:1,The expression of VEGF-D,VEGFR-3 were significantly higher in lymph node metastasis group than in no lymph node metastasis group; the result reveals that VEGF-D and its receptor VEGFR-3 play a key role in breast tumor lymph node metastasis. 2,In the primary infiltrating ductal carcinoma, the immunostaining of VEGFR-3 was not only localized in the interstitical vessel endothelial cell cytoplasm, but also in the carcinomotous cytoplasm; The expression of VEGF-D was low in breast benign tissue, but it was very high in breast cancer; it suggests that breast cancer cell may product VEGF-D.3,The VEGFR-3 positive intersittical vessel may be the principle pathway in tumor invasion and metastasis.
