BackgroundAccording to WHO's statistics, lung cancer has already become the first threat of cancer killer to human life. Seeing from the types of lung cancer tissue, the incidence of lung adenocarcinoma was only next to the incidence of squamous carcinoma, and the data statistics of recent years indicate that its incidence trend to be obviously increased as well. Local invasion and hematogenous metastasis of adenocarcinoma of lung are ofen earlier than those of squamous cell carcinoma which at the first position, while the malignant biological behavior such as invasion and metastasis are currently the biggest obstacle to the treatment of adenocarcinoma of lung, resulting in its clinical therapeutic efficacy and prognosis badder than squamous cell carcinoma. Tumor invasion and metastasis are the multi-step complex processes, including tumor cell defluvium, migration, adhesion, degradation of extracellular matrix, angiogenesis, etc., with a in-depth study, in the above process, especially in the process blood vessel neogenesis, the study of looking for targeted therapy point to preventing tumor invasion and diversion of received extensive attention.ETS'family belongs to proto-oncogene, the members encoding outputs constitute a large family of transcriptional control factor, participating those functions such as control cell proliferation, differentiation, migration, apoptosis, angiogenesis and organ formation etc. Ets-1 (E26 tansformation-specific 1) is a representative transcription factor in the family. Existing studies indicate that, Ets-1 was overexpressed in a variety invasive tumors of human being, while was not expressed in normal differentiated tissue, which hints that Ets-1 plays an important role in the evolution of malignant tumors. A further study found that Ets-1 may be through transcriptional activation the expression of a series of genes which play an important role in altering the extracellular matrix and angiogenesis, such as matrix metalloproteinases(MMPs), urokinase plasminogen activator(uPA)and so on, participating extracellular matrix degradation, cell migration, tumor angiogenesis, invasion and metastasis and many other areas. It is reported in the literature that Ets-1 and vascular endothelial growth factor (VEGF) have an intimate relationship in the process of infiltration and metastasis as well as angiogenesis in noumenal tumor.ObjectiveTo detect Ets-1 and VEGF protein expression level in lung adenocarcinoma tissue and far-adjacent normal lung tissue, analyze their dependablity with clinical pathological data such as degree of tumorcell differentiation, lymph node metastasis, clinical stage, and study the correlation of Ets-1 and VEGF to the occurrence, development and clinical features of lung adenocarcinoma.MethodsIn this study, immunohistochemical staining (SP method) was used to detect the protein expression of Ets-1 and VEGF in 46 cases of lung adenocarcinoma and 16 cases of adjacent normal tissue (both were archived wax blocks of surgical removal tissue). We also analyze the correlation between them, and explore their clinical significance. All specimens were pathologically confirmed lung cancer tissue or normal lung tissue.ResultsEts-1 and VEGF had different levels of expression in 46 cases of lung adenocarcinoma tissue: Ets-1 positive expression rate was 71.74% (33/46), VEGF positive rate was 76.09% (35/46). Both were obviously higher than them in16 cases of adjacent normal tissue:,12.50% (2 / 16) and 25.00% (4 / 16), the difference had statistically significant (P <0.05). In lung adenocarcinoma tissues, following with the average diameter increasing, degradedness of tumor differentiation degree and lymph node metastasis, the positive expression of Ets-1 was increased, and the difference was statistically significant (P <0.05); The positive expression of VEGF closely related to size of tumor, lymph node metastasis and TNM stage( P <0.05); the protein expression of Ets-1 and VEGF was positively correlated in lung adenocarcinoma(P <0.05).ConclusionsThe expression of Ets-1 and VEGF in lung adenocarcinoma tissues was obviously higher than that in adjacent normal lung tissue, positive expression of Ets-1 and VEGF was correlated with the process of invasion and metastasis as well as angiogenesis of lung adenocarcinoma. The detection of Ets-1 and VEGF in lung adenocarcinoma tissue will help to comprehend the malignant biological behavior of lung adenocarcinoma, and they may be used to assess the development and prognosis of lung adenocarcinoma.
