The Effect Of Jujuboside A On Acute Ocular Hypertension- Induced Damage In Rat Retinal Ganglion Cells

Objective To establish acute ocular hypertension model and detect the protection effect of jujuboside A on retinal ganglion cells(RGCs)in SD rats with acute ocular hypertension.Methods1,60 SD rats were divided randomly into control group, one day model group,three day model group, five day model group,seven day model group.Except control group,the rat model with acute ocular hypertension was established by increasing intraocular pressure through anterior chamber infusion. IOP was measured using the Tono-Pen before,after and immediately in surgery and then at two day's interval.Morphology was observed before or after the surgery. The levels of mRNA of THY-1 in every group were detected by RT-PCR.2,48 SD rats were divided randomly into control group,model group and jujuboside A group.Except control group,the rat model with acute ocular hypertension was established by increasing intraocular pressure through anterior chamber infusion. After the establishment of model,SD rate were injected with jujuboside A by tail vein(20mg/kg/d,7days),model group was injected with physiological saline replaced of the same volume jujuboside A.Changes in histological retina and the number of RGCs were observed by immunohistochemistry. The levels of m RNA and protein of THY-1 were detected by RT-PCR,western blot.Results1,IOP were15.80±1.47mmHg at control group.The IOP of every model group in surgery was 30.13±2.36mmHg.The IOP returned to normal at 3,5,7day after reperfusion.2,With time pass by after the modeling, THY-1mRNA expression and Positive cells of ganglion cell layers gradually reduced.3,Retinal THY-1 expressed in the ganglion cell and inner plexiform layers by immunohistochemistry. 7days after model established,the number of THY-1 positive cells in model group is reduced compared to that in control group(P <0.01).No statistical difference in that between Jujuboside A and control group.4,After transient ischemia, the levels of total retinal Thy-1 mRNA and protein in model group were dramatically reduced compared to that in control group by RT-PCR and western blot (P<0.01). No statistical difference in that between Jujuboside A and control group.Conclusion:1,To establish acute high intraocular pressure rate mould through anterior chamber infusion can obtain pathology change that is similar to acute angle-closure glaucoma.2,Jujuboside A have an effect on rats with acute ocular hypertension by protecting retinal ganglion cells.
