PURPOSE To observe aortic atherosclerosis and its relationship with serum levels of CD40/CD40L, CD40/CD40L expression of aortic tissue in rats model induced by the combined high-salt and high-fat diet and effects of statins in the processMETHOD 1. A total 60 healthy SD male rats, rats aged 8 weeks, weight 200-250 g, were randomly divided into five groups (12 in each group):N group (normal control group), S group (high salt group), F group (high fat group), SF group (high fat high salt compound group), T group (high salt and high fat+simvastatin).2. In each group were fed with different feed. S: standard diet; S group:high-salt diet; F group:High fat diet; SF Group:high-salt diet fat compound; T group:fat combined with high salt diet+Simvastatin.3. After 10 weeks feeding with rat tail blood pressure (noninvasive) measurement of arterial blood pressure of rats in each group; 4. Abdominal aorta blood, enzyme-linked immunosorbent assay to determine serum concentrations of CD40/CD40 L; 5. Take 1 initial segment of the aortic root, HE staining of frozen sections of aortic atherosclerosis and the inflammation response to the situation; 6. Immunohistochemical detection of aortic root vessel wall middle group CD40/CD40L expression, image analysis was used to analyze pathological Photo slice analysis system (positive cells), to reflect the strength of CD40/CD40L expression.RESULTS (1) Blood pressure:S, F, SF, T blood pressure (systolic and diastolic blood pressure) than N group (normal control group), the difference was statistically significant (p<0.05). SF group than in S, F group increased, the difference was significant (p<0.05), T less than the SF group in blood pressure, and S, F group difference was not statistically significant (p>0.05).(2) Arterial pathology:SF group continuity in rat aortic endothelial damage, partial wall to the lumen, forming plaque, plaque formation within the necrotic material and elastic fibers rupture, arterial wall thinning of tear crack, was more typical atherosclerotic plaque changes, inflammatory cells infiltration; F group also appears atherosclerosis change, but the severity lighter than the SF group; S, T group showed membrane thickening, not See atherosclerosis; N group no abnormal changes.(3)Serum sCD40L levels:S, F, SF serum sCD40L levels were higher than the N, T group, the difference was significant (p<0.05); SF group than in S, F group, the difference was statistically significant (p<0.05); S group and between group F, N group and T group no significant differences between (p> 0.05).(4) Srterial tissue CD40/CD40L expression:S group, F group, SF Group artery tissue CD40/CD40L expression was higher than N, T group, the difference was significant (p<0.05); SF group than in S, F group, the difference was statistically significant (p<0.05), S, F group no significant differences between (p>0.05); N, T was no significant difference between the groups (p> 0.05). CONCLUSION (1) A significantly elevation of blood pressure can be iuduced by high salt diet, but the change is enhanced by combined high-salt with high fat diet.(2) High fat diet can conduce aortic atherosclerosis,and high salt ingestion exaggerate the result, which probably is engaged by inflammatory process, and CD40/CD40 L overexpression participate in pathological ways.(3) Simvastatin protect the vascular from irritant by inhibiting possibly the serum and arterial tissue expression of CD40/CD40 L.
