Object: This study was to examine mRNA expressions of Toll-like receptor-4,Humanβ-defensin-2 and Tumor necrosis factor-αin placenta tissue of preeclampsia and control group, and to explore the differences and relationships among them, so as to provide the significance basis for the pathogenesis of preeclampsia.Method: Placental beds were obtained from 65 women in the following groups: (1) women with preeclampsia (n=45, mild=20, Severe=25); (2) women in control group (n=20). We examined the messenger RNA (mRNA) of TLR-4, HBD-2and TNF-αin placenta using real-time reverse transcription polymerase chain reactions by dsDNA-binding dyes of SYBR Green, which calculated the relative expression using 2-△△Ct method.Result: (1) Women of two groups had expressed TLR-4mRNA,HBD-2mRNA and TNF-αmRNA; (2)The mRNA expressions of TLR-4,HBD-2 and TNF-αwere significantly higher in preeclampsia than in control group (p<0.01, p<0.05, p<0.05 respectively), and increased by the state of illness aggravating and worsening, however, there was no significantly statistical difference between mild and severe in preeclampsia (p>0.05); (3)There showed a significant linear positive correlation between TLR-4 and TNF-αin preeclampsia only(r=0.832, p<0.05), not in control group (r=0.19, p>0.05). Whereas other relationships did not exist (p>0.05).Conclusion: Increased expression of TLR-4,HBD-2 and TNF-αmay play an important role in the pathogenesis of preeclampsia, which TLR-4 and HBD-2 maybe mediated by increased expression of TNF-αto participate in the pathophysiology of preeclampsia.
