Objective:To investigate the association between nonsyndromic cleft lip with or without cleft palate (NSCL/P) and genetic polymorphisms of Wnt-5a/rs566926 in Gansu province of China.Methods:Genomic DNA was isolated from peripheral lymphocytes of 200 patients with NSCL/P and 200 normal match controls. According to Wnt-5a/rs566926 gene polymorphism design a primer, Wnt-5a genotypes were determined by polymerase chain reaction (PCR). Wnt-5a/rs566926 variant was detected by DHPLC. We performed the case-control analysis and used transmitted disequilibrium test (TDT) to calculate 22 heterozygous parents with an affected child. The variants in the NSCL/P patients with and without family history were examined byχ2 analysis.Results:In case-control analysis, there were significant differences in C allele genotype of Wnt-5a/rs566926 among NSCL/P compared with the control group (χ2=22.753, P<0.05), A allele frequency was 60%, and C allele frequency was 40%in the patients, A allele frequency was 75.75%, and C allele frequency was24.25%in the controls. There was no Genetic imbalance in the 22 heterozygous parents with an affected child during transmission disequilibrium test (χ2= 1.504, P>0.05). The frequency was no significance between the cases with and without family history (P>0.05).Conclusions:We found that there are genetic polymorphisms of Wnt-5a/rs566926 in the people of Gansu province of China, and it may be a risk factor for NSCL/P in Gansu province. There was no statistical significance between the cases with and without family history, but further research is warranted with a larger sample size.
