| Objective 1.To semi-quantitative detect DNAJB6 mRNA and DNAJB6 protein expression in human breast cancer;2.The relationship between the expression of DNAJB6 mRNA and DNAJB6 protein with the clinicopathologic features was analyzed.3.The relationship between the expression of DNAJB6 mRNA with DNAJB6 protein was analyzed.Methods Eighty five pairs samples of breast cancer and adjacent normal tissue were obtained1.Total RNA was extracted and mRNA expression of DNAJB6 was proceeded by real-time fluorescence quantitative RT-PCR;2.The protein expression of DNAJB6 in human breast cancer was measured immunohistochemically;3. The relationship between the expression of DNAJB6 mRNA and DNAJB6 protein with the clinicopathologic features of human breast cancer was investigated.Results 1.Of the total 85 pairs of sample,52 pairs of the DNAJB6 expression was down-regulated(61.2%) ,and down-regulated expression in breast cancer was associa -ed with the size of the tumor , TNM staging and histological grade (p<0.05);2.The rate of low expression of DNAJB6 is 61.2% (52/85),and the low expression in brea -st cancer was associated with the size of the tumor and histological grade(p<0.05).3. The expression of DNAJB6 mRNA has correlated with DNAJB6 protein (t=13.983,p<0.001).Conclution 1.The expression of DNAJB6 mRNA and DNAJB6 protein in human breast cancer were down-regulated;2. The low expression of DNAJB6 might be a biological indicator of high-degree of malignantcy and poor prognosis. |
