By studying the expression of O6-Methylguanine-DNA-Methyltransferase (MGMT), Glutathione-transferase pi (GST-π) and TopoisomeraseⅡ(TopoⅡ) in human gliomas cell with survival time of patients, as well as the relations among the three in glioma, in order to judge the prognosis of patients, and give guidance to the plan of chemotherapy.59 specimens of gliomas were collected from patients who underwent surgeries at the Affiliated Hospital of Hebei University. All tissues were routinely fixed in 10% formalin, dehydrated conventionally, embedded in paraffin wax, and then archived for later use. Serial sections of 5μm were cut from the blocks. Then representative sections were stained with hematoxylin and eosin(H&E)in order to confirm the histopathological type; Investigate The surial time of the patients; The expression of MGMT,GST-πand TopoⅡwere detected by using En Vision immunohistochemical method; Semi-quantitative analysis was made on the results of the immunohistochemical SP staining according to Krajewska method; All the statistical analyses were made with SPSS17.0 for Windows software.χ2 test was used in counting non-ranked data,and Spearman correlation rank test were performed in analysis of correlation, Meanwhile, survival time of patients with MGMT,GST- and TopoⅡexpress or not draw a Kaplan-Meier survival curves and log-rank test. For all the tests, results with P values lower than 0.05 (P<0.05)are considered statistically significant. Results:The expression of MGMT in human gliomas exists heterogeneity,Statistically there are no significant differences among MGMT expressions in gliomas of different grades (P>0.05) the expression rates of GST- and TopoⅡwere obviously different statistically among patients of different pathological grades (P<0.05);Patients with MGMT,GST-πand TopoⅡexpression showed a shorter survival time than those with no MGMT,GST-πand TopoⅡexpression (P<0.05); No correlation was found between the positive expressions of any two of the three express in gliomas (P>0.05)The expression of MGMT in human gliomas exists heterogeneity,and the expression of MGMT has nothing to do with the grade of glioma; Expressions of GST-πand TopoⅡare indexes in judging the degree of malignancy of gliomas; Expressions of MGMT,GST-πand TopoⅡare indexes in judging the prognosis of patients; Several drug resistance genes co-express, and checking the expressions of the genes together can provide a prospective guidance to clinic drug use for glioma patients undergoing chemotherapy.
