ObjectiveBy immunohistochemical (IHC) immunohistochemistry methods, detection the expression of ERCC1 and MGMT in normal brain tissues and different levels of gliomas, And analyzes its Correlation. Offer reference for the development of clinical individual chemotherapy.MethodsBy immunohistochemistry,respectively detection the expression of ERCC1 and MGMT in 41 cases of different levels glioma and 10 cases of normal brain tissue. And analysis the correlation of them.Results1.19 patients with gliomas of 41 were positive with MGMT. The overall positive expression rate is 46.34%; The positive rate of expression of different levels as follows:the positive rate of grade I glioma was 36.37%(4/11); The positive rate of Gradeâ…¡glioma was 40%(4/10); The positive rate of Gradeâ…¢glioma was 54.55% (6/11);The positive rate of gradeâ…£was 55.56%(5/9).MGMT expression rate increased with the rise of Glioma pathological level.By statistical analysis, the different of different levels of glioma was no statistically significant, (P>0.05)2.21 patients with gliomas of 41 were positive with ERCC1. The overall positive expression rate is 51.22%. The positive rate of expression of different levels as follows:the positive rate of grade I glioma was 27.27% in (3/11);The positive rate of gradeâ…¡glioma was 40.00%(4/10);The positive rate of gradeâ…¢glioma was 63.64%(7/11); The positive rate of grade IV was 77.78%(7/9).ERCC1 expression rate increased with the rise of Glioma pathological level.By statistical analysis,the different of different levels of glioma existing statistical significance.3.10 cases of cerebral hemorrhage in the resection line decompression surgery within the normal brain tissues of MGMT and ERCC1 expression was negative or weakly positive. The expression of MGMT and ERCC1 were significantly different(P<0.05) compare with Glioma tissue.4. MGMT and ERCC1 expression in glioma cells was correlated and there was positively correlated between them.There were no significant difference With gender and age of the expression of MGMT and ERCC1 in the 41 patients'glioma organization.Conclusions1. MGMT expression in gliomas of different levels was of different, and its expression rate increased with the rise level of gliomas, the higher Glioma levels the higher relatively expression of MGMT.But, statistical analysis the difference was not statistically significant.explain that MGMT expression and tumor grade had no correlation.2. ERCCl expression in gliomas of different levels was of different, and its expression rate increased obviously with the rise level of gliomas, the different expression exist statistical significance. The expression intensity was positively correlated with glioma grade. explain that MGMT expression was related with tumor grade.3. In glioma, the expressions of MGMT and ERCC1 were no relative with age and sex. In clinical diagnosis and treatment process,sex and age can not be used as a reference factor when Setting individualized chemotherapy for patients.4. MGMT and ERCC1 expression in glioma cells was correlated and there was positively correlated between them. This suggests that two genes in the gliomas occurrence,development,growth and malignant invasion play a mutual adjustment, mutual coordination effect. Can be used as a index of prognosis monitoring tumor recurrence or metastasis. Detection of the two genes would some reference to the Clinical treatment of glioma. 5. The expression of MGMT and ERCC1 in normal brain tissues was negative or weak positive.
