Ginger, as the perennial herb belonging to zingerberaceae, is the traditional material which is both spice and traditional Chinese medicine. In this paper, essential oil and gingerol extraction,isolation, purification and assay process conditions have been studied.By steam distillation and SDE have been used to extact ginger essential oil, the rates were 2.15% and 2.82%. The components separated were identified by GC/MS.The results showed essential oil of ginger could be extracted effectively, 36 and 43 essential compounds were identified by the two methods respectively.They composed of 91.54%and 93.49% of the total essential oil respectively. The relative percentages of the constituents were determined by normalization method.Two methods of extracting oil, basically the same composition mainly terpenes compounds. In comparison,rates of SDE is higher than steam distillation,and used shorter time. Therefore, SDE is better than the method steam distillation.Extracted by SDE, main components are essential terpenes compounds (67.73%), contain alcohols (21.04%), aldehyde ketone (3.59%) and other compounds (1.13%). The highest content was Zingiberene (18.99%), followed byβ-Sesquiphellandrene (14.35%),α-Farnesene (9.95%),α-Curcumene (8.77%) andβ-Bisabolene (5.63%).The five components of all was detected, the composition of 57.69% preliminary determination for ginger oil main chemical compositions.Then respectively extracted through such methods as the refluxing method, microwave-assisted extraction and ultrasonic extraction auxiliary method of ginger powder of gingerol extraction, the ultrasonic extraction auxiliary extraction gives the highest extraction percentage, achieve the highest 7.28 mg/g. The optimum extraction conditions showed that the solvent-solid ratio1:13.7,ultrasonic time 25.9min,ethanol concentration 90.0%.The predicted gingerol extraction yield was 7.38mg/g ,and the verification yield was 7.41mg/g.Gingerol was separated and purified using polyamide resins for chromatographic column packing. Static test shows, polyamide resin for gingerol saturated the adsorption quantity for 8.34 mg/g, adsorption desorption rate 75.8%,psychotic rate range was 52.2~78.9%,Dynamic test on the optimum technological conditions for 160mL samples 2mL/min,75% of the ethanol concentration as eluent, dosage for 180mL, in these conditions, gingerol purity achieved 53.6%.Using a quick method-High Performance Capillary Electrophoresis(HPCE) to analysis ingredient 6-gingerol of ginger.Specific separation condition of buffer for 10mMNa2HPO4-30mM borax-30% (Φ) acetonitrile (pH=9.5), for 280nm detected wavelength,using 0.5 psi pressure test sample in time for the sample and 5s,capillary column temperature of 20℃, separation voltage for 15 kV electrophoresis. In the condition ,the content of 6-gingerol in ethanol extract was 0.32 mg/mL.
