Objective:This study was to observe the early renal pathological changes of rabbits given high lipid diet and the effects of using Gypenosides.Methods:50 male rabbits,10 to normal group, the rest 40 were given high lipid diet of the group.Testing the TC,TG content in blood serum of the 50 rabbits after four weeks. Rabbits with TC content more than 6 times of normal group rabbit were successful model rabbits. Eliminating successful model rabbits with weak constitution, choosing 30 successful model rabbits, which were continued to be given high lipid diet for another four weeks.Dividing the 30 rabbits randomly to 3 groups, the model group, the simvastatin group and the Gypenodide group. After another four weeks testing thet blood serum TC, TG content, renal pathological changes and detecting the renal Desmin alpha-SMA, TGFβ1, VCAM-1 protein expression of the 40 rabbits.Results:1.After given the high lipid for eight weeks, the blood creatinine and the blood urea nitrogen of the four groups are the same.2.Model group rabbits'mesangial cells were slightly hyperplastic.3 (1) Compared to normal group, model group Desmin (p < 0.001), alpha SMA (p< 0.001), VCAM-1 (p< 0.001) were significantly higher. TGF-β1 expression had no statistically significant differences compared to normal group.(2)Compared to model group, simvastatin group Desmin, alpha SMA expression were significantly lower (p< 0.01 Desmin; alpha SMA, p< 0.001), TGF-beta expression had no statistical significant differences compared to model group.VCAM-1 expression was significantly lower than the model group (p< 0.01).(3)Compared to the model group, the Gypenodide group Desmin, alpha SMA expression were significantly below the model group (p< 0.001 Desmin; alpha SMA, p< 0.001); TGF-β1 expression had no significant differences compared to model group; VCAM-1 expression were significantly below the model group (p< 0.01).Conclusion:1.Lipid abnormity can induced some cell factors and protein of rabbit kidney expression abnormally and damage the kidney finally.2 Gypenodide can protect kidney by reducing the abnormal expression of cell factors induced by lipid abnormity. The mechanism can relate to its effects on high lipid and inflammation reation.
