| Objective:explore and discuss the treatment mechanism of Bai Tou Weng Decoction in curing inflammatory bowl disease (IBD) of the acute period through measuring MPO level and expression of NF-κBp65 and p50 after conducting the IBD model of the rats by coloclitis with TNBS.Methods:Divide 40 male Wistar rats into normal control group, model group and low dose group of Bai Tou Weng Decoction, high dose group of Bai Tou Weng Decotion and Mesalazine group, and apply corresponding chinese formular or western medicine to different groups separately except the normal control group and the model group with the 0.9% physiological saline after 24- hour- coloclitis -molding process with 75mg/kg dosage of TNBS. Give all of the groups'intragastric administration once a day for 14 days, observe diarrhea and hemafecia condition and record weight changes simultaneously.16 days after model establishment put all the rats to execution and draw materials which the experiment needs. Measure CPR (C Reactive Protein) of arteria femoralis and MPO (Myeloperoxidase) level of the colon tissue, and observe the gene expression of NF-κBp65 and p50 protein by SABC, Western Blotting and PCR methods.Results:compaired with the control group, the histological score of colon mucous membranes and MPO activities of other groups are both higher (P< 0.05), while the histological score of colon mucous membranes and MPO activities of other groups are lower when compaired with the model group (P<0.05). C Reactive Protein has no statistics significance. The NF-KBp65 and mRNA expression of colon tissue of model group is higher than normal control group, while the NF-KBp50 and mRNA expression is lower than it. The NF-KBp65 and mRNA expression of high dose group of Bai Tou Weng Decoction is lower than that of the model group, while the NF-KBp65 and mRNA expression of low dose group of Bai Tou Weng Decoction is higher than that of model group. But the NF-KBp50 and mRNA expressiones of both the high dose and low dose group are higher than that of model group. And NF-KBp65 and p50 expression of the posotive medicine group is reduced.Conclusion:The high dose group of Bai Tou Weng Decoction can reduce the NF-KBp65 and mRNA expression and increase the NF-KBp50 and mRNA expression simultaneously. It is may ane of the treatment machanisms. But it is different from the effect machanism of 5-aminosalicylic acid (5-ASA). |
