Tumor Drug Sensitivity Test And Its Clinical Significance

Objective:To explore Cancer susceptibility testing in vitro of six kinds of malignant solid tumor cells and its benefit for clinical individualized chemotherapy from the individuals,the histogenesis,and the pathologic types and so on.Methods:Analyzing results of chemosensitivity test by MTT for 494 patients with 5 kinds of malignant tumor in 1991 to 2006 and the relationship with clinical therapy retrospectively. Also observed the long-term survival using survival analysis.Results:Six kinds of solid tumor cells are of different chemotherapy drugs showing different sensitivity but for lymphoma (P<0.05).Five kinds of solid tumor cells, sensitivity rate of DDP, colorectal cancer is higher than other four kinds, the liver cancer is higher than other four kinds in ADM sensitivity rate , the sensitivity rate of Vp-16, ovarian cancer is relatively higher(P<0.05). Three kinds of digestive system malignant tumor, colorectal cancer have significantly higher sensitivity rate in DDP,MMC,5-FU,the sensitivity rate of ADM and VP– 16 Liver cancer is significantly higher(P<0.05). Between pathological for low differentiated adenocarcinoma, colorectal cancer showed higher sensitivity rate in DDP than other three kinds of low differentiated adenocarcinoma, similarly,ovarian cancer showed higher sensitivity rate in ADM, and lung cancer showed higher sensitivity rate in HCPT and Vp-16,the sensitivity rate in TPT have no statistically significant. Susceptibility about stomach cancer, lung cancer and ovarian cancer have no statistically significant between the various pathological classification (P>0.05). Also,susceptibility in stomach cancer,livercancer and ovarian cancer have no statistically significant too between various clinical stages ,except lung cancer,ADM,VCR,GEM showed different chemosensitivity in different clinical phases(P <0.05), and the relatively higher sensitivity is stageⅡcases. About the 3-year survival of gastric cancer patients,3-year survival of lung cancer patients,5-year survival of ovarian cancer patients, the sensitive group were higher than the control group,but there was no statistically significant, 5-year survival of liver patients was lower than control group, the difference was no statistically significant too.And In COX proportional hazard model analysis about four kinds of malignant solid tumors, MTT susceptibility test is not a significant and independent prognostic factor.Conclusion:The same tumor of different individuals,5 kinds of solid tumors,3 kinds of digestive system tumors,different low differentiated adenocarcinoma all showed different chemosensitivity in vitro. Between different solid tumor pathological types and different clinical stages did not differ in the drug sensitivity.Different individuals and the different tumors have different cancer drugs sensitivity,MTT have a good correlation with clinical chemotherapy drugs and is helpful to screen effective chemotherapy drugs,and eliminate unsensitive drugs,and beneficial to choose effective clinical chemotherapy. However,susceptibility testing is not yet a significant factor of survival.
