Objective: To analyze the differences of the cell cycle proteins cyclinD1,p16,p27 in blood lymphocyte in gastric cancer patients pre-and post-operation and further explore the effect of perioperative intervention therapy with elemene on these cell cycle proteins.Methods: 48 cases gastric cancer patients who were newly diagnosed and prepared operation, were randomly divided into two groups, one group is the adjuvant treatment group with elemene in perioperative period, one group is the surgery alone group. Flow cytometry was used to detect the expression of the cyclinD1,p16,p27 in blood lymphocyte pre-and post-operation of the intervention group and the surgery alone group. SPSS13.0 statistical package were used for statistical analysis, results to mean±standard deviation, the differences of cyclinD1,p16,p27 between the two groups were compared by using two independent samples t or t'test,there was significant difference (p <0.05).Results: In the surgery alone group, the expression of cyclinD1,p16,p27 in peripheral blood lymphocyte before and after surgery were significantly differen(tp<0.05),among the expression of cyclinD1,p16 after surgery were lower than that before surgery, the expression of p27 after surgery was higher than that before surgery; in the intervention group, the expression of cyclinD1,p27 in peripheral blood lymphocyte were significantly different before and after surgery(p<0.05),among the expression of cyclinD1 after surgery were lower than that before surgery, the expression of p27 after surgery was higher than before surgery, but the expression of p16 before and after surgery had no significant difference (p> 0.05); the expression of cyclinD1,p16 after surgery between intervention group and the surgery alone group were significantly different(p<0.05),in which the expression of cyclinD1 in intervention group after surgery were lower than that in the surgery alone group, the expression of p16 in intervention group after surgery was higher than that in the surgery alone group; the expression of p27 after surgery between intervention group and the surgery alone group had no significant difference (p> 0.05). Conclusion: The expression of cyclinD1,p16 in peripheral blood lymphocyte in surgery alone group reduced, this expression pattern may be the recurrence and metastasis model of advanced gastric cancer. The perioperative adjuvant treatment with elemene can accelerate the reduction rate of cyclinD1 in peripheral blood lymphocytes, inhibit the decrease of the expression of p16. So the perioperative adjuvant treatment with elemene ,this mechanism may curb the recurrence and / or metastasis of advanced gastric cancer patients whom hard surgical excision. There is no response on expression of p27 in peripheral blood lymphocytes of advanced gastric cancer after perioperative intervention treatment with elemene.
