| Objective:study on the quality of commodity specifications and grades in gastrodia by detection of physical chemistry and sensory evaluation,to discuss the correlation of product specifications and grades with the indexs of quality evaluation,the sientific rationality of traditional standard of product specifications and provide agents on the basis of new industry standards.Methods:Survey and typical sampling to determine collecting product specifications grades sample of Gastrodia;Reference 2005 edition of Pharmacopoeia of The People's Republic of China,to content detection and statistical analysis of the moisture,water extraction,alcohol-soluble extraction,total ash in Gastrodia;To optimize extraction of gastrodia polysaccharide by the single factor and orthogonal design;The determination of Gastrodin and P-hydroxybenzyl alcohol in Gastrodia by using High Performance Liquid Chromatography(HPLC)and statistical analysis;Gastrodia HPLC fingerprints established based on optimization parameters fingerprint index F;Cluster analysis and discriminant analysis were used in recognition research on Gastrodia different product specifications grades;The evaluation index system of Gastrodia commodity specification was established for the first time,which based on the delphi method.Results:1.The content of moisture in Gastrodia ranged from10.61%to 21.50%,59% of the samples exceeded failure;Total ash content range is 1.57~4.59%,97% of the sample compliance. 2.Water-soluble extract concentration range is 5.41-64.38%,95% of the samples is greater than15%;with75% ethanol as extraction solvent of alcohol-soluble extract,alcohol-soluble extract concentration range is 9.98~29.55%,97% of the samples is greater than 10%.3.Water extract and alcohol precipita optimum conditions:extraction temperature 100℃,extraction time 4h,feed water ratio1:40;the content of gastrodia polysaccharide ranged from8.15%to62.13%,89% of the samples is greater than 14%.5.Gastrodin content range is 0.195~1.144%,97% of the samples gastrodin content(≥0.20%)qualified;P-Hydroxybenzyl alcohol content range is 0.015~0.248%,95% of the samples is more than 0.02%.6.Based on the indexs of quality evaluation to conducte related analysis and component analysis,the results the indexs related to each other and with certain regularity(P<0.05),but with specifications,rank correlation is not strong or inconsistent with the law;Testing targets only the source of the sample producing a certain relevance,established a recognition model of authentic or non-authentic(P<0.05)based on the logistic regression,the predicted correct rate is 80%.7.Gastrodia fingerprint cluster analysis and discriminant analysis:the region is divided into authentic or non-authentic,the origin is divided into four,the grades is divided into two,the rate of discrimination and the clustering results of classification in line is up to 79.2%,87.5%,90.5%,the classification model effect is good.8.Delphi method experts were active(the recovery=84.2%,100%),reliability (the coefficients of authority was0.8313,0.8583),and the coordination between the experts(Kendall"W=0.220,0.278,P<0.05),and the product specification grade evaluation system of gastrodia was established.Conclusion:The indexs related observably to each other(P<0.05)and with certain regularity,but with specifications and grades correlation is not strong or inconsistent.The Region,Origin,Harvest Period,Cultivated or Wild,Grade, all is the important factors of the quality of Gastrodia.The industry standards of gastrodia commodity specifications grades is scientific,reasonable,feasi-ble by authentic or non-authentic,different regions or origins,different harvest period,cultivated or wild and the gastrodia counts of unit weight. |
