Objective:On the basis of determining the effective dose of Astragalosides (AST) and Panax Notoginseng Saponins (PNS) in anti-cerebral ischemia model in mice,explore AST and PNS in compatibility.And explore AST and PNS compatibility on oxidative stress,energy metabolism and matrix metalloproteinase of early cerebral ischemic referfusion injury in mice.Method:1. Filled the stomachs of the mice with different dose of AST and PNS at preestablish. Cerebral ischemia in mice were prepared by bilateral common carotid artery and vagus ligation,and record the time of resistant ischemia. To determine the AST and PNS effective dose of anti-cerebral ischemia and effective dose in compatibility.2. C57BL/6N mice were randomly divided into sham-operation group, model group, AST and PNS compatibility of high, medium and low-dose group, AST group, PNS group and Edaravone group. Cerebral ischemia-reperfusion injury were prepared by bilateral common carotid artery ligation for 20 min followed by 60 min reperfusion after administration for 4 days. Observe the contents of Adenosine Triphosphate(ATP),Adenosine Diphosphate (ADP),Adenosine Monophosphate (AMP),Malonaldehyde (MDA),Glutathione (GSH),Nitric Oxide (NO), total adenine nucleotides(TAN), energy charge(EC) and activity of Superoxide Dismutase (SOD),Nitric Oxide Synthase (NOS) in brain tissue after reperfusion. Analyze whether AST has an interaction of compatibility with PNS.3. C57BL/6N mice were randomly divided into sham-operation group, model group, AST and PNS compatibility of high, medium and low-dose group, AST group, PNS group and Edaravone group. Cerebral ischemia-reperfusion injury were prepared by bilateral common carotid artery ligation for 20 min followed by 24h reperfusion after administration for 4 days. Observe the expression of MMP-9 and TIMP-1 protein in brain tissue after reperfusion. Analyze whether AST has an interaction of compatibility with PNS.Conclusion:1. Work out the dose of compatibility is AST110+PNS 115mg/kg in anti-cerebral ischemia-reperfusion injury.2. Compared with sham-operated group, the content of MDA,NO and the activity of NOS in model group's brain tissue increased remarkably (P <0.01), the activity of SOD,Na+-K+-ATPase and the contents of GSH,ATP,TAN and EC were decreased(P<0.01). Drug groups significantly inhibited the increase in MDA,NO,NOD and the decrease in SOD,GSH,Na+-K+-ATPase,ATP,TAN,EC in brain after cerebral ischemia-reperfusion (P<0.01~P<0.05). The compatibility effect of AST110mg/kg and PNS115mg/kg is more obvious.AST110mg/kg and PNS 115mg/kg compatibility has some interactions (P<0.01~P< 0.05).They showed synergy on increasing the activity of SOD and the content of ATP in the brain tissue, and showed additive action on reducing the content of MDA,NO and the activity of NOS and increasing the contents of TAN,GSH,EC and the activityof Na+-K+-ATPase.3. Compared with sham-operated group, MMP-9 in model group's brain tissue increased remarkably (P<0.01),TIMP-1 have have no changes (P>0.05).Drug groups significantly inhibited the increase in MMP-9 and the decrease in TIMP-1 in brain after cerebral ischemia-reperfusion (P<0.01~P<0.05). The compatibility effect of AST110 mg/kg and PNS115mg/kg is more obvious.AST110mg/kg and PNS115mg/kg compatibility has some interactions(P<0.01~P<0.05). They showed additive action on reducing the MMP-9 and TIMP-9 increasing activity.Conclusion:1. Effective com-ponent from Chinese medicinal herb can produce noticeable results.2.The compatibility of the AST and the PNS have inhibited effect in the obstacle of energy metabolism and oxidative stress injury,and can adjust the expression of MMP-9 and TIMP-1 protein in brain tissue after cerebral ischemia, and the compatibility between the AST and the PNS has a synergistic or additive effect.
