Association Research Between Polymorphism Of IFN-γ And IL-10, Environmental Risk Factors, And Susceptibility To Esophageal Cancer | | Posted on:2011-02-28 | Degree:Master | Type:Thesis | | Country:China | Candidate:W P Du | Full Text:PDF | | GTID:2154330332970325 | Subject:Epidemiology and Health Statistics | | Abstract/Summary: | PDF Full Text Request | | ObjectiveTo explore the polymorphism of Interferon-γ(IFN-γ) and Interleukin-10(IL-10) with susceptibility to esophageal cancer, as well as the interacti-onbetween the environmental risk factors and the polymorphism of IFN-γor IL-10.To explore the interaction between the polymorphism of IFN-γand IL-10.To explore IFN-γexpression in the serum and the relationship with esopha-geal cancer.MethodPCR technology was used to determine the genotypes of IFN-γ+T874A and IL-10 G1082A;Enzyme linked immunosorbent assay method was used to detect IFN-γ;All subjects were investigated with same questionnaire.Statistics analysis was carried out by SAS 9.0 and SPSS13.0 statistical package withχ2-test,t-test and logistic regression analysis.The gene-gene and gene-environment interaction on EC risk was evaluated by the crossover analysis.Result1.All esophageal carcinoma case patients and control subjects were adequately matched by age and sex.2.IFN-γ+T874A have three kinds of genotype which were AA,AT and TT.Frequencies of them were 32.5%,59.17% and 8.33% in case group,and 46.7%,49.2%,4.1% in control group respectively(χ2=5.9492,P=0.0511); Allele frequencies of T were 37.92% in case group higher than 28.7% in control group (χ2=4.6414,P=0.0312);Compared with IFN-γ+874AA genotype, OR(95%CI) of IFN-γ+874AT genotype was 1.729(1.015~2.947) and OR(95%CI) of IFN-γ+874AT+TT genotype was 1.821 (1.081~3.069)IL-10-G1082A have three kinds of genotype which were AA,AG and GG. Frequencies of them were 80.51%,16.95% and 2.54% in case group, and 86.6%,12.6%,0.8% in control group respectively(χ2=2.1770,P=0.3367); The G allele frequency of IL-10 gene in the case group was not higher compared with that in the control group(χ2=2.1561,P=0.1420).Compared with IL-10-1082AA genotype, OR(95%CI) of IL-10-1082AG+GG genotype was 1.559(0.777~3.127).3.Analysis of multiple logistic regression showed that drink index,liking to eat hot and hard diet,moldy food,regular diet habit, fresh vegetables,family history of tumor,manner in deal with things and mental status were the main environmental risk factors of esophageal carcinoma. Their OR respectively were 1.669(95%CI:1.242~2.243),2.333(95%CI:1.540~3.533),3.699(95%CI:1.275~10. 731),3.524(95%CI:.1.941~6.401),2.027(95%GI:1.321~3.109),27.661(95%CI:5.1 28~149.211),1.684(95%GI:1.131~2.507),1.992(95%CI:1.060~3.744),2.012(95 %CI:1.236~3.221).4.Interaction analysis results:according to the addition model, there was synergistic action between IFN-γ+874AT+TT and drink index,moldy food and regular diet habit, their S respectively were 1.909,4.154 and 5.026, and there was synergistic action between IL-10-G1082A and attitude in dealing with affairs and mental status, their S respectively were 4.519,9.64.5.The interaction between IFN-γ+874AT+TT and IL-10-1082AG+GG was not increased the risk of EC OR=2.342 (95%CI:0.975~5.626)6.Density of IFN-γin blood serum in case group and control group was not statistically significant differences (t=1.73, P=0.0886)Conclusion1.The IFN-γ+874AT genotype might be a risk factor to EC.2.Drinking,liking to eat hot and hard diet,mildewed food,fresh vegeta-bles,regular diet habit,family history of tumor,manner in deal with things and mental status were the main environmental risk factors of esophageal carcinoma in Dawen River basin.3.There existed synergistic interaction between IFN-γ+874AT+TT genotype and environmental factors such as drinking,moldy food and regular diet habit.And synergistic interaction between IL-10-1082AG+GG genotype and environmental factors such as manner in deal with things and mental status were found. 4.IFN-γ+874AT+TT genotype and IL-10-1082AG+GG genotype had no synergistic action in interaction. | | Keywords/Search Tags: | esophageal carcinoma, IFN-γ, IL-10, environmental factors, interaction, case-control study | PDF Full Text Request | Related items |
