Lotus (Nelumbo nuxifera Gaerth) leaves was a traditional Chinese medicine for treatment of summer heat, clear yang and hemostasia. The clinic medicine research found that lotus leaves extract had strong bioactivity ingredients, including flavonoid and alkaloid and so on, which had reducing blood lipid, antibacteria, antivirus, antioxidation and some other activities. Pharmacological research about lotus leaves mainly focused on losing weight and lowering blood lipid, and it was rarely reported the anti-bacterial and anti-inflammatory activities and pointed out the pure compound. Gaining pure compound with anti-bacterial and anti-inflammatory activities as a goal, separation and purification of the lotus leaves crude extract were taken, while testing the anti-bacterial and anti-inflammatory activities of fractions from each step. Structure analysis of pure compound was carried on in this paper, which could be a reference for the lotus leaves research and development in antibacteria and antiinflammation.Lotus leaves were decocted in hot water with a little ethanol and tested the extract's antibacterial and anti-inflammatory activities. It was showed that the lotus crude extract had excellent antibacterial and antiinflammtory activities, whose inhibition against S. mutans and clearance against IL-6 and IL-8 were more than 95%. Crude extract was isolated by resin column and eluted with distilled water and 20%, 50%, absolute ethanol. Isolates eluted by distilled water and 20%, 50% ethanol had bad antibacterial and antiinflammtory activities, whose inhibition against S.mutans and clearance against IL-6 and IL-8 were less than 30% and 50% separately. Isolate eluted by absolute ethanol had good antibacterial and antiinflammtory activities, whose inhibition against S.mutans and clearance against IL-6 and IL-8 were more than 90%. The isolate eluted by absolute ethanol was divided into chloroform and methanol two groups by solubility. The fractions separated by silica gel show different antibacterial and antiinflammtory activities. Fractions from chloroform and methanol showed good antibacterial activity and good anti-inflammatory activity separately. Fractions with small polarity and medium polarity had the best antibacterial activities; fractions with medium and large polarity had the best activities in clearing IL-8; fractions with minimum polarity and medium polarity had the best activities in clearing IL-6. It was showed that antibacterial and antiinflammtory activities of lotus leaves were due to various substances and antiinflammtory mechanisms were also different. Because the fractions with medium polarity show the good antibacterial and antiinflammtory activities, it was purified by silica gel and gained a pure compound named substance A, whose MIC50 was less than 5mg/ml and MIC90 was 0.04mg/ml against S.mutans, A.a, P.g, Candida and the minimum concentration with antiinflammation was 0.2mg/ml. As the result of'H-NMR,13C-NMR,'H-'H COSY, HMQC, MS, UV, IR, it is identified that the substance A was lysicamine which was separated from lotus leaves firstly. |
