Objective:To study the protective effect of mangiferin on daunorubicin-induced rat cardiomyocytes peroxidation and the possible mechanism.Methods:Create cardiomyocytes injury model through 4μmol/L DNR,then treat with different concentrations of Mangiferin to detect cardiomyocytes viability by MTT assay;to detect LDH,CKand CK-MB by Automatic biochemical analyzer;to detect the content of SOD and MDA by Spectrophotometer, to detect the level ROS andΔΨm by FCM; determine Hsp70 and Catalase CAT gene expression change by fluorescence quantitative PCR.Results:1.After the injury of DNR,Cardiomyocyte gradually shrink ,even some beat disorder or stop beating. with time. The high and middle mangiferin group can relieve the Cardiomyocyte injury caused by DNR,while the VitC group's protection was not obvious.2.Cardiomyocyte livability was gradually up with the concentration of mangiferin up,the high and middle mangiferin group have obvious protection on cardiomyocyte.Compaired with DNR model group and VitC group, 100μmol/L mangiferin group can improve cardiomyocyte viability obviously(P<0.01).3. After 48 hours of DNR added into medium ,the content of LDH,CK,CK-MB in medium increased much. With the mangiferin concentration up,the leakage of Enzyme.There is statistical differences between 100μmol/L mangiferin group and DNR model group (P<0.01).4. After 48 hours of DNR added into medium, the content of MAD in medium raised much,while the activity of SOD went down. Mangiferin can reduce reduce MDA production through Antioxidant and increase SOD activity.5. After 48 hours of DNR added into medium, the content of ROS in Cardiomyocyte increased obviously. With the mangiferin concentration up, the content of ROS in Cardiomyocyte decreased gradually,the middle and high mangiferin group can clean up the ROS in Cardiomyocyte powerfully. Compaired with DNR model group and VitC group, 100μmol/L mangiferin group can decrease the ROS in Cardiomyocyte.6. After 48 hours of DNR added into medium, theΔΨm decreased much. With the mangiferin concentration up, theΔΨm in Cardiomyocyte gradually restored. Compaired with DNR model group and VitC group, 100μmol/L mangiferin group can restoreΔΨm obviously,while there is no difference between VitC group and DNR model group(P=0.368).7. After 48 hours of DNR added into medium, the expression of HSP70 and CAT gene in Cardiomyocyte increased. With the mangiferin concentration up, the expression of HSP70 and CAT gene in Cardiomyocyte decreased. Compaired with DNR model group, 100μmol/L mangiferin group can decrease the expression of HSP70 and CAT gene.Conclusions: In vitro, Mangiferin can relieve myocardial toxicity induced by daunorubicin, the possible mechanism is related with mangiferin can effectively clear the ROS and relieve peroxidation.
